- Previous reports have described a spectrum of clinical phenotypes in patients with pathogenic variants. One specific phenotype is the Basel-Vanagaite-Smirin-Yosef syndrome, which is a rare, autosomal recessive disorder characterized by a wide range of symptoms including eye abnormalities, cardiac abnormalities, palatal abnormalities, intellectual disability, and epilepsy. Although epilepsy appears to be common, the electroclinical phenotypes of patients with Basel-Vanagaite-Smirin-Yosef syndrome have not been well described. Here, we report 3 Basel-Vanagaite-Smirin-Yosef syndrome patients from 2 families of Lebanese descent with a homozygous founder variant from Alberta Children's Hospital, Calgary, Alberta. We describe in detail electroclinical phenotypes in each of these 3 patients. These patients developed seizures with onset between 2 and 3 years of age, multifocal and generalized discharges, as well as photoparoxysmal responses on electroencephalogram. Ng Andy Cheuk-HimD'Alfonso SabrinaInnes A MicheilScantlebury Morris H - Respiratory syncytial virus (RSV), the most common cause of bronchiolitis and pneumonia in infants, elicits a remarkably weak innate immune response. This is partly due to type I interferon (IFN) antagonism by the non-structural RSV NS1 protein. It was recently suggested that NS1 could modulate host transcription via an interaction with the MED25 subunit of the Mediator complex. Previous work emphasized the role of the NS1 C-terminal helix α3 for recruitment of the MED25 ACID domain, a target of transcription factors (TFs). Here we show that the NS1 α/β core domain binds to MED25 ACID and acts cooperatively with NS1 α3 to achieve nanomolar affinity. The strong interaction is rationalized by the dual NS1 binding site on MED25 ACID predicted by AlphaFold and confirmed by NMR, which overlaps with the two canonical binding interfaces of TF transactivation domains. Single amino acid substitutions in the NS1 α/β domain, notably NS1 E110A, significantly reduced the affinity of NS1 for MED25 ACID, both in vitro and in cellula. These mutations resulted in attenuated replication of recombinant RSV (rRSV-mCherry). They did not significantly upregulate type I or III IFN levels in IFN-competent BEAS-2B cells, contrary to the NS1 α3 deletion. However, in line with attenuated replication, the NS1 E110A mutation enhanced expression of the antiviral interferon-stimulated gene ISG15, and NS1 I54A upregulated ISG15, OAS1A and IFIT1 in IFN-competent cells. In MED25-knockdown cells, rRSV-mCherry replication was further attenuated at a late post-infection timepoint. The difference between WT and NS1 mutant rRSV-mCherry was partially lost, suggesting that the NS1-MED25 ACID complex contributes to controlling antiviral responses at this timepoint. The strong interaction and the extended binding interface between NS1 and MED25 ACID provide evidence for a mechanism, where NS1 blocks access of transcription factors to MED25, and thereby MED25-mediated transcription activation. - Source: PubMed

Recent scientific advances have highlighted the SCF-JAZ-MYC2-MED25 module as a central hub for JA signaling, orchestrating transcriptional repression, derepression, activation, amplification, and feedback termination. This review summarizes current insights into the roles of JA in the regulation of biotic and abiotic stress responses and agronomic traits, including root development, regeneration, fertility, flowering, leaf senescence, and seed development, with a particular emphasis on the crosstalk between JA and a wound-induced peptide hormone, systemin, which mediates systemic wound responses. A deeper understanding of the JA regulatory mechanisms will provide valuable strategies for engineering crops with enhanced stress resilience and improved yields. Li YanbingWu FangmingLi Chuanyou - The pigment glands of cotton synthesize the toxic gossypol that contributes to cotton's defence, but also limits the usage of cottonseed as a food source. Previous studies have shown that the gland-localized GoPGF is the master transcription factor controlling the gland morphogenesis and gossypol biosynthesis in cotton. However, the precise mechanism underlying the GoPGF-mediated transcription of downstream genes remains unclear. In this study, TurboID-based proximity labelling was established in cotton using a transient expression system to identify the components of GoPGF-complex. A total of 48 high-confidence GoPGF-proximal proteins were identified, and a mediator subunit, MED25, was shown to control gland formation through phenotypic studies. Several lines of evidence indicate that GhMED25 physically interacts with GoPGF in the nucleus, and GhMED25-GoPGF complex regulates the expression of GhJUB1 related to gland morphogenesis, and several well-characterized gossypol biosynthetic genes. ChIP-qPCR further revealed that GhMED25 acts as an enhancer of the GoPGF-mediated transcriptional output. Our study shows the value and reliability of the proximity labelling system in studying the low-abundance and cell-type-specific proteins in crops, adding a new role to the multifunctionality of MED25 in plants, and provides new insights into the intricate signalling network governing gland formation and terpene production in cotton. - Source: PubMed
