- The decay B^{±}→K_{S}^{0}π^{±}, with a CP asymmetry expected to be close to zero in the standard model, is theoretically clean and sensitive to potential new physics. An analysis of the decays B^{±}→K_{S}^{0}π^{±} and B^{±}→K_{S}^{0}K^{±} is performed using proton-proton collision data collected by the LHCb experiment at a center-of-mass energy of 13 TeV, corresponding to an integrated luminosity of 5.4 fb^{-1}. The CP asymmetries are determined to be A^{CP}(B^{±}→K_{S}^{0}π^{±})=-0.028±0.009±0.009 and A^{CP}(B^{±}→K_{S}^{0}K^{±})=0.118±0.062±0.031, and the branching fraction ratio is measured to be B(B^{±}→K_{S}^{0}K^{±})/B(B^{±}→K_{S}^{0}π^{±})=0.055±0.004±0.002, where the first uncertainties are statistical and the second are systematic. These results are the most precise measurements of these quantities to date. A search for the rare decay B_{c}^{±}→K_{S}^{0}K^{±} is also performed. No significant signal is observed, and the upper limit on the product of the branching fraction ratio B(B_{c}^{±}→K_{S}^{0}K^{±})/B(B^{±}→K_{S}^{0}π^{±}) and the fragmentation-fraction ratio f_{c}/f_{u} is set to be 0.015 (0.016) at the 90% (95%) confidence level. - Source: PubMed

pathological feature of systemic sclerosis (SSc), but its immune regulatory mechanisms remain incompletely understood. In particular, the expression patterns, clinical relevance, and functional roles of CD8CD28 and CD8CD28 angiogenic T cell (Tang) subpopulations in SSc have not been fully defined. This study aimed to systematically investigate the distribution, clinical significance, and molecular characteristics of these Tang subsets in SSc. We identified CD8 Tang subpopulations in the lung tissue of patients with SSc-associated interstitial lung disease (SSc-ILD), with a gradual decrease in CD28-related signals along the inferred differentiation trajectory. In the peripheral blood of patients with SSc, CD8CD28 Tang were increased, whereas CD8CD28 Tang were decreased. These alterations were associated with vascular complications, including ILD, digital ulcers, and advanced microvascular lesions, as well as with disease activity, and may serve as biomarkers of treatment response. Functional experiments showed that CD8CD28 Tang impaired endothelial cell proliferation, migration, tube formation, and survival through a GZMB-dependent mechanism associated with reduced Akt activation, whereas CD8CD28 Tang exerted protective effects through VEGF/AREG-related activation of EGFR/Erk signaling. In summary, our data support an association between CD8CD28/CD8CD28 Tang imbalance and clinical parameters of vasculopathy in SSc, while in vitro findings indicate distinct endothelial-injurious and endothelial-protective functions of these two subsets, suggesting that they may contribute to vascular dysfunction in SSc. - Source: PubMed

However, data regarding the effect of the human lung environment on γδ T cell functionality remains limited. To address whether lung inflammation impacts γδ T cell functionality, we analyzed lung and matched hilar lymph node (LN) tissue from deceased donors and patients with interstitial lung disease (ILD). We performed high-parameter spectral flow cytometry to examine the expression pattern of phenotypic biomarkers and assess ex vivo function. We identified lung-specific enrichment of γδ T cells with an effector memory phenotype relative to matched regional LN. We then used an ex vivo stimulation approach to interrogate the capacity to protect against infection (granzyme B [GzmB], interferon-γ [IFNγ] and tumor necrosis factor [TNFα]) and promote epithelial cell proliferation (amphiregulin [AREG]). We found that γδ T cells in lung and LN from deceased donors had similar functional properties. While γδ T cell populations from ILD lungs largely maintained cytokine production capacity, expression was diminished relative to LN counterparts. Importantly, lung γδ T cells maintained polyfunctional GzmB, IFNγ and TNFα expression across cohorts.
