- Objective To investigate the effect of gentiopicroside on osteogenic differentiation of human bone marrow mesenchymal stem cells (BMSCs), and to determine whether its mechanism involves the stromal cell-derived factor 1(SDF-1)/C-X-C chemokine receptor 4 (CXCR4) pathway. Methods BMSCs were divided into six groups: normal culture control group, osteogenic induction model group, low-dose gentiopicroside (L-gentiopicroside, 10 μmol/L) group, medium-dose gentiopicroside (M-gentiopicroside, 20 μmol/L) group, high-dose gentiopicroside (H-gentiopicroside, 40 μmol/L) group, and H-gentiopicroside+SDF-1/CXCR4 pathway inhibitor (AMD3100) group (H-gentiopicroside+AMD3100, 40 μmol/L gentiopicroside+10 μg/mL AMD3100). Cell viability, apoptosis, ALP activity, mineralized nodule formation, and protein levels of the SDF-1/CXCR4 pathway were assessed using the CCK-8 assay, flow cytometry, ALP staining, Alizarin Red S staining, and Western blotting, respectively. Results No mineralized nodules were observed in either the control and model group, although the color of the model group deepened. Compared with the control group, the model group showed significantly increased A value, ALP activity, expression levels of Runt related transcription factor 2 (RUNX2), osteopontin (OPN), SDF-1, CXCR4 proteins, along with a lower apoptosis rate. - Theaflavin 3,3'-digallate (TF3), a flavonoid compound, exerts antidiabetic effects, induces apoptosis, and suppresses cancer growth. However, its effects on melanoma metastasis and insulin-like growth factor 1 (IGF-1)-induced cancer stemness remain largely unexplored. This study explored key molecular signaling pathways underlying the antitumor and antimetastatic activities of TF3 in melanoma. The antimetastatic potential of TF3, as well as its impact on IGF-1-induced cancer stemness, were evaluated using wound healing assays, gelatin zymography, Matrigel invasion assays, sphere formation assays, and Western blotting. Its antiangiogenic and antimetastatic activities were further examined in vivo using subcutaneous xenograft and tail vein injection mouse models. TF3 significantly inhibited cell migration, invasion, and matrix metalloproteinase (MMP) activity in A 375 and A2058 melanoma cells. It also suppressed sphere formation, self-renewal capacity, and aldehyde dehydrogenase 1 (ALDH1) activity. In sphere-forming melanoma cells, TF3 downregulated key cancer stemness and drug resistance markers, including ABCB1, ABCG2, CD44, and CXCR4. Notably, TF3 reversed IGF-1-induced sphere formation, invasion, and MMP-9 expression. Mechanistically, TF3 suppressed IGF-1-mediated phosphorylation of focal adhesion kinase (FAK) and Src. In vivo, TF3 significantly inhibited tumor growth, reduced angiogenic marker expression, and suppressed lung metastasis. Although serum B-cell maturation antigen (sBCMA) has been shown to be a prognostic and monitoring tool for multiple myeloma patients, it has not been evaluated among those with WM. We assessed sBCMA levels among 50 WM patients: 16 had received prior therapy, while 34 were treatment-naïve (TN), 19 of whom never went on to require therapy (smoldering [S]WM). Levels were higher among WM patients (median, 63.76 ng/mL) than healthy subjects (median, 35.24 ng/mL; p < 0.0001). TNWM patients who required therapy showed higher initial levels (median, 137.71 ng/mL) than those with SWM (median, 50.19 ng/mL; p = 0.0002). sBCMA also correlated with disease status: levels decreased at best response (≥ minimal response; median, 38.36 ng/mL) compared to baseline (median, 117.00 ng/mL; p = 0.0078) and increased among patients who developed progressive disease (median, 79.42 ng/mL; p = 0.0625). Baseline sBCMA levels positively correlated with β2 microglobulin (p = 0.0193) and M-protein (p = 0.0450), and negatively with hemoglobin (p = 0.0090) and albumin (p = 0.0016). Overall, these findings suggest sBCMA may serve as a biomarker for prognosis and monitoring for WM patients. - Source: PubMed
