- Microbial chain elongation for medium-chain fatty acid production has gained significant attention. This study elucidated how initial pH and carbon sources regulate microbial caproic acid production from organic waste. By successively subculturing pit mud with Huangshui, lactic acid, or ethanol at different pH values (4.0-6.0), we obtained two enriched consortia: NatCom H (from Huangshui) and NatCom E (from ethanol), both of which produce 10 g/L caproic acid at pH 6.0. Key functional divergence was observed: NatCom H maintained production from pH 5.0 to 6.0, whereas NatCom E was functional only at pH 6.0. Integrated multiomics and bioreactor analyses revealed that this divergence stemmed from distinct microbial interactions. NatCom H was structured around a pH-mediated mutualism between Clostridium tyrobutyricum and acid-sensitive Caproiciproducens species, engaging in sequential substrate conversion. In contrast, NatCom E was dominated solely by Clostridium kluyveri. The hypothesized interaction mechanism was confirmed by isolating the key strains (including two novel Caproiciproducens species) and constructing a synthetic community, which demonstrated that C. tyrobutyricum alleviates acidic stress for Caproiciproducens, enabling caproate synthesis at pH 5.0. These findings reveal a substrate-driven ecological strategy for environmental adaptation and provide a mechanistic framework for engineering robust consortia to convert wastes such as Huangshui into valuable caproic acid. - Source: PubMed

Despite their advantages over other nanomaterials that include controlled spatial presentation of targeting ligands such as lipids and sugars, understanding their cell targeting and uptake mechanisms remains limited. Here, we investigated NANP cellular targeting, uptake, and delivery of small interfering RNAs (siRNAs) to liver and neuronal cell models . Using a rational design approach, we targeted NANPs to two clinically validated receptors, the asialoglycoprotein receptor (ASGPR) and the low-density lipoprotein receptor (LDLR), respectively, using GalNAc and lipidation. We systematically evaluated how the ligand valency, interligand spacing, linker length, and ligand chemistry affected NANP association with on- and off-target liver cell types, revealing the relative roles of the biomolecular corona, receptor engagement, and endocytosis in these targeting strategies. We found that lipidation enhanced NANP uptake into HepG2 cells, a model cell line for hepatocytes, by promoting apolipoprotein recruitment, LDLR engagement, and clathrin-mediated endocytosis and also increased association with nonparenchymal cells. HepG2 uptake was further improved by conjugating NANPs to lipids with higher valency provided that lipids were adequately displayed away from the surface of NANP edges with more lipophilic lipids yielding greater cell association. We then benchmarked the potential for NANPs to deliver siRNAs to HepG2 cells in comparison with lipid nanoparticle and conjugate technologies and explored lipid functionalization as a strategy for nonhepatic NANP targeting to model neuronal cells. Clonal evolution analysis reveals no dominant RT-resistant clones, prompting a genome-wide CRISPR screen to identify radiosensitizing targets. The screening highlights DNA damage response genes, validating the effectiveness of our approach. N-acylneuraminate-9-phosphatase (NANP), a critical enzyme in the sialic acid synthetic pathway, is top-ranked in the screening and associated with patient outcomes. After radiation, NANP-deficient cells exhibit more DNA damage, G2/M arrest and apoptosis, and impaired DNA repair by favoring non-homologous end-joining over homologous recombination. Mechanistically, NANP influences NF-κB signaling and the mesenchymal state by modulating sialylation and internalization of tumor necrosis factor receptor 1 (TNFR1), thereby affecting RT sensitivity. Intracranial orthotopic xenograft experiments validate the function of NANP in vivo. As protection is understood to be antibody-mediated for both vaccines, the success of a multistage vaccination strategy will be aided by understanding factors that impact the cellular drivers of humoral immunity, e.g. dosing regimen, exposure to malaria, and age. Prior analyses of adult vaccinees showed higher RH5-specific memory B cell responses with delayed fractional versus monthly booster dosing (50-50-10μg at 0-1-6months and 10-10-10μg at 0-1-2months, respectively), but the B cell impact of delayed booster regimens has not yet been evaluated in the target paediatric population. We introduce a nucleic acid nanoparticle (NANP)-based platform designed to engage with the human immune system in a controllable manner. By varying the chemical composition while keeping the architectural parameters constant, we identified key factors that influence the immunorecognition and stability of NANPs. Chemical modifications provide additive functional regulation, while pairing NANPs with different delivery agents further enhances their immunomodulatory capabilities. Specific compositions boost T cell effector functions, promote B cell proliferation, and activate innate immune populations in human peripheral blood mononuclear cells. The resulting interferon responses and immune signatures mimic those triggered by viral infections. Notably, in vivo studies demonstrate that NANPs function as efficiently as traditional clinically used adjuvants, without increasing the risk of autoimmunity. This suggests that NANPs are a safe and effective tool for immunotherapy with promise for developing formulations targeting cancer and infectious diseases. - Source: PubMed
