We describe a nontoxic, scalable, and environmentally sustainable ovine cartilage decellularization protocol utilizing supercritical CO(scCO). Five scCO decellularization protocols were evaluated in various ovine cartilage dimensions, of which the multicycle scCO method was selected for evaluation of ovine zested cartilage, ovine minced cartilage, and human minced cartilage. Cartilage groups were enclosed in porous polylactic acid (PLA) scaffolds and implanted subcutaneously on rat dorsa along with an empty scaffold control group. Rats were euthanized after 1, 3, and 6 months for volumetric, histological, and biomechanical analysis of implants. The efficiencies of scCO decellularization was enhanced by multicycle process or additional reagents, as well as the smaller dimensions of processed cartilage, as confirmed by DNA quantification and nucleic staining. Preserved structural integrity was evidenced by intact cartilage structure, lacunae components, and collagenous content postprocessing on histology. Implantation in a murine model for 6 months demonstrated excellent neo-tissue ingrowth with minimal inflammatory response. Biomechanical testing revealed no significant differences in moduli or hydraulic permeability of minced and zested samples, before and after the decellularization treatment. The resulting stiffness after implantation mimicked the physical properties of other cartilaginous tissues. In conclusion, we demonstrate the effective decellularization of ovine cartilage graft with a scCO protocol while maintaining native tissue architecture and biomechanical properties with a minimal immunogenic response . This study introduces a comprehensive, accelerometer-based approach to examining real-world UE use, including the contribution of UE activity during walking, and compares UE use during walking versus ADL-based activities. People with subacute stroke wore bilateral wrist and ankle inertial measurement units (IMU) for one week (24 h/day). Agreement between UE-Total, UE-Continuous Walking and UE-ADL measures were assessed using Bland-Altman analyses. Variables were modeled using linear regression to explore step count, mobility aid use and UE motor impairment as predictors of IMU measures of real-world UE use. Agreement between UE-Total and UE-ADL measures showed minimal bias. Differences between UE-Continuous Walking and UE-ADL varied by mobility aid. UE-Continuous Walking was associated with increased UE activity and relatively greater paretic arm use compared with UE-ADL. Across UE-Continuous Walking and UE-ADL, step count was the strongest predictor of real-world UE use. UE activity during continuous walking exhibits different use patterns as compared to ADL-based activity. Accordingly, separating continuous walking-related UE activity from ADL-based UE use may improve clinical interpretation of IMU measures of real-world UE use in individuals with subacute stroke. - Source: PubMed

However, the discontinuous nature of recycled fibres requires efficient alignment and consolidation. In this study, highly aligned discontinuous carbon fibre tapes were manufactured from recycled carbon staple fibres and bio-based polyamide 11 (PA11) fibres using a textile-based processing route and compression moulding. Two material systems containing nominal fibre mass fractions of 50 wt.% and 70 wt.% rCF were investigated. The resulting laminates were characterised in terms of fibre volume content (FVC), fibre orientation distribution, and mechanical performance. FVC of up to 56.72 vol.% were achieved for the 70 wt.% rCF material. Tensile testing revealed a significant increase in stiffness and strength with increasing fibre content, reaching values of 48.1 GPa and 866 MPa. The longitudinal compression modulus and the in-plane shear modulus showed similar trends, while the transverse tensile strength and compressive strength remained strongly influenced by local defects and fibre-matrix interactions. A 2D mesoscopic image analysis demonstrated a pronounced preferential fibre orientation, with approximately 69% of the analysed fibre regions aligned within ±10° of the dominant fibre direction. The derived alignment coefficient η correlated well with the tensile modulus, confirming the strong influence of mesoscopic fibre architecture on mechanical performance. The results demonstrate that the combination of bio-based PA11 and oriented rCF, enabled through textile-based alignment technologies, provides a promising pathway towards sustainable lightweight composite structures. - Source: PubMed
