- KIF21A, a member of the Kinesin-4 family of motor proteins, is involved in the regulation of microtubule dynamics and intracellular transport, with emerging evidence suggesting its potential role in cancer progression. In this study, we performed an integrative analysis of over 3825 human phosphoproteomics studies to characterize site-specific phosphorylation of KIF21A. Three predominant phosphosites were identified in KIF21A (S853, S1212, and S1239) with the highest detection frequency across phosphoproteomics studies, and were analyzed for co-regulation patterns to identify potential kinase associations and functional networks. Phosphosite S853 showed a strong association with cytoskeletal organization and cortical microtubule stabilization complexes (CMSCs) components, including KANK1 (S186), PHLDB2 (S513, S42) and CLASP1 (S600, S572, S646), indicating its role in cytoskeletal organization. Upstream kinase analysis identified potential regulators, such as PAK2, RPS6KA1/A3, RPS6KB1, CHEK1/2 and CDK18/16 with site-specific variability in their associations with KIF21A predominant sites. Interestingly, phosphosite-specific correlation analysis between KIF21A and candidate kinases revealed that the KIF21A S1239 phosphosite exhibited tumor-specific correlations with CDK18 across multiple cancer types. Functional enrichment revealed that co-regulated phosphoproteins were involved in cytoskeleton regulation, cell cycle regulation, and carcinogenesis. Pan-cancer analysis demonstrated dysregulated expression of KIF21A in multiple tumor types, with stage-associated upregulation in selected cancers. Gene-level validation further supported these findings, showing consistent positive correlations between KIF21A and key regulators such as CTNND1 and PTK2, as well as other cytoskeleton and cancer-associated genes. Overall, this study highlights site-specific phosphorylation as a key regulatory mechanism of KIF21A and suggests its involvement in cytoskeleton-associated signaling networks in cancer. - Source: PubMed

Neutrophil-lineage cells occupy a unique position in this setting because they are generated, retained, mobilized, aged, and reprogrammed within the same bone marrow niches that disseminated tumor cells exploit for homing and survival. This review examines how neutrophils, tumor-associated neutrophils, immature neutrophils, low-density neutrophils, and PMN-MDSCs shape skeletal colonization. We discuss tumor-to-marrow signaling, CXCR2-dependent recruitment, CXCR4/CXCL12-mediated marrow retention, neutrophil-circulating tumor cell interactions, vascular arrest, dormancy escape, NET-mediated matrix remodeling, immune suppression, and effects on osteoclast-osteoblast coupling. Evidence is strongest in breast and prostate cancer models, where pathways such as CXCL5/CXCR2, CTNND1-CXCR4/CXCL12, PR3-RAGE, and DKK1-CKAP4-STAT6-CHI3L3 link neutrophil-lineage cells to skeletal progression and immunotherapy resistance. However, several mechanisms, including CTC-neutrophil clustering and NET-driven dormancy awakening, remain partly extrapolated from non-skeletal models. We therefore emphasize evidence hierarchy, methodological limitations, and therapeutic opportunities, arguing that selective reprogramming or functional inhibition of pro-metastatic neutrophil states may be more promising than indiscriminate neutrophil depletion in metastatic bone disease. A clearer understanding of these context-dependent neutrophil programs may help refine biomarker development and guide combination therapies for patients with skeletal metastases. - Source: PubMed

Cleft palate only (CPO) is an OFC subtype. Here, we searched for novel nsCPO risk genes carrying homozygous and compound-heterozygous variants, by analyzing exome data from six sibling pairs with nsCPO born to unaffected parents. After stringent quality control and filtering, we identified 6 homozygous variants and 32 compound-heterozygous variants in 5 and 16 candidate genes, respectively. We prioritized DDR2, a collagen-activated receptor-tyrosine-kinase influencing extracellular matrix composition, as our top candidate for functional follow-up, since variants in this gene can cause Warburg-Cinotti syndrome, the phenotypic spectrum of which includes palatal abnormalities. Knock-down and knock-out of DDR2-orthologs in zebrafish caused craniofacial abnormalities resembling CPO in humans. Zebrafish immunostaining indicated that DDR2-orthologs were expressed in mature head muscle cells, while murine single-cell RNA-Sequencing data detected Ddr2 expression only in head muscle progenitor cells; the latter finding was confirmed in human embryo sections stained for DDR2. DDR2-expressing head muscle progenitor cells may influence extracellular matrix composition through DDR2-mediated signaling, thereby affecting outgrowth, elevation, and fusion of the palatal shelves, a previously postulated mechanism involved in palatogenesis. Most established OFC genes (e.g. CDH1, CTNND1, IRF6, and GRHL3) act via mechanisms related to epithelial integrity and periderm differentiation, whereas our data provide evidence supporting DDR2 as a risk gene for nsOFC that functions by influencing extracellular matrix composition. - Source: PubMed

Traditional submerged cell cultures lack airway-like polarity and structural complexity, limiting studies. This study utilized a porcine air-liquid interface porcine respiratory epithelial cell (ALI-PREC) model to investigate infection dynamics, cytopathic effects (CPEs), and host-specific responses. Primary tracheal epithelial cells isolated from three 6-week-old pigs were cultured under air-liquid interface conditions for 4 weeks to form differentiated pseudostratified, ciliated epithelium. ALI-PRECs were inoculated with strain 232 at 10, 10, and 10 CCU/mL for 2 or 5 h and monitored for 144 h. Quantitative microscopy assessed total particle count, area, average size, and percentage area covered by particles, normalized against mock controls. adhered within 2 h, inducing CPE (cell rounding, clustering, and detachment) and reducing ciliary activity in a dose- and time-dependent manner. DNA was detected in the epithelium by 24 h post-inoculation (hpi) and in subnatants at higher doses, demonstrating epithelial barrier disruption. Ciliary beating persisted in some replicates, suggesting interindividual variability in host responses. Friis medium had no detectable impact on ALI-PRECs, validating its suitability for infection studies. Gene expression analysis revealed downregulation of ciliary motility genes (, , , and ) at 72 hpi and upregulation of intercellular junction genes (, , and ) by 120 hpi, suggesting a wound healing response. The ALI-PREC model effectively mirrors key aspects of pathogenesis, providing a robust platform for studying host-pathogen interactions and identifying therapeutic targets.IMPORTANCEDespite its economic impact on pigs, the mechanisms by which () causes enzootic pneumonia remain poorly understood. Early infection events (bacterial exposure and adherence) are hard to study because traditional cell cultures lack airway complexity and cellular diversity. We developed an model replicating porcine airway structure and cell populations. Using this model, we showed that rapidly adheres to airway cells and disrupts ciliary function and epithelial integrity in a dose- and time-dependent manner. These effects coincide with altered expression of genes governing ciliary motility and cell-cell junctions. Importantly, our model revealed natural variability in cytopathic responses among pigs, indicating that host-specific factors influence disease progression.
