Although most ALS cases occur sporadically, without a known family history of the disease, genetic factors play a major role in its pathogenesis through monogenic, oligogenic, or polygenic mechanisms. It is estimated that 10-15% of ALS cases occur in a familial setting; however, a specific monogenic cause cannot always be identified. Establishing the underlying genetic basis in both sporadic and familial ALS is essential, as it enables individualized and family genetic counseling, facilitates the early identification of at-risk or oligosymptomatic relatives, improves the prediction of gene-specific clinical trajectories, and, more recently, determines eligibility for gene-targeted therapies, such as tofersen for -associated ALS and ulefnersen, currently under clinical investigation, for -associated ALS. Over the years, differing opinions have existed regarding the role of genetic testing in individuals diagnosed with ALS. However, accumulating clinical evidence has increasingly supported the timely and early implementation of genetic testing as part of the standard clinical management of patients with ALS. In this study, acute toxicity of the single and combined pollution of Polyethylene microplastics (PE MPs) and diesel on Tegillarca granosa after 96 hours exposure was investigated in terms of oxidative stress related biochemical indicators, histopathology, DNA damage and gene expression, respectively. The results showed that microplastics-diesel combined exposure resulted in increased SOD activity in both the gills and the visceral mass of T. granosa, whereas GPx and GST activities were inhibited. However, no significant change in MDA content was observed under short-term exposure conditions. The Integrated Biological Responses version 2 (IBRv2) index revealed that the gills exhibited higher sensitivity to the combined pollutant. High-concentration combined exposure induced histopathological alterations (gill swelling, digestive tubule necrosis of the visceral mass) and DNA damage. Furthermore, high-concentration combined pollution upregulated the gene expression of cytochrome P450 Family 2 (CYP2 family), GST, CAT, GPX, and SOD1, while downregulating the KEAP1. It indicated that combined pollutants can regulate antioxidant genes by activating the cytochrome P450 (CYPs) enzyme system and the KEAP1-NRF2 signaling pathway, interfere with the antioxidant defense system of T. granosa, and produce an enhanced toxic effect. The research revealed the toxic effects and molecular mechanisms of microplastic-petroleum combined pollution on T. granosa, providing a scientific basis for assessing the ecological risks of combined pollution in marine environments. - Source: PubMed

This study evaluated the impacts of environmentally relevant concentrations (100 and 1000 ng L⁻¹) of cannabinol, codeine, and methamphetamine, individually and in mixtures, on the growth and development of Crassostrea virginica larvae. Eight-day exposures significantly reduced larval survival, impaired growth, and altered swimming behavior, with mixtures producing the strongest effects. At 1000 ng L⁻¹ , cumulative survival reached zero in mixture and cannabinol groups, while methamphetamine and codeine reduced survival by 74.8% and 56.0%, respectively. Drug mixtures also caused the greatest reduction in larval growth (13.3%) and mean swimming velocity (67.5%) relative to controls, with significant shifts from predominantly rectilinear swimming to circular or motionless behavior. Short-term (24 h) exposures induced changes in the expression of stress-related genes (mapk14, hsp70, sod1, and gst), with hsp70 exhibiting the strongest response (14- to 39-fold upregulation), indicating rapid activation of oxidative stress and detoxification pathways, particularly under mixture conditions. These findings demonstrate that DAs, especially in combination, can disrupt physiological and molecular processes in oyster larvae at concentrations reported in surface waters. Given their persistence, increasing consumption, and documented occurrence near wastewater discharges, DAs warrant consideration as contaminants of emerging concern in coastal ecosystems and should be incorporated into ecological risk assessments that account for mixture effects and early-life stage sensitivity. - Source: PubMed
