Adedoyin Festus FataiAlola Andrew AdewaleBekun Festus Victor - Perfluoroalkyl substances (PFAS) are persistent synthetic chemical compounds that are widely distributed in the environment and pose significant risks to both human and wildlife. In this study, the seasonal distribution of PFAS was investigated in water and sediment samples collected from four major Rivers in Korea: the Nakdong, Geum, Seomjin, and Yeongsan Rivers. To assess bioaccumulation, medaka fish (Oryzias latipes), a freshwater bioindicator, were simultaneously collected at six natural field habitats (Yeosu, Gimhae, Naju, Andong, Gunsan, Dangjin). The concentrations of ∑PFAS in River water samples ranged from of 4.71 ~ 372.24 ng/L in the Geum River (median = 56.72), 2.51 to 158.91 ng/L in the Yeongsan River (median = 27.35), 0.44 to 247.63 ng/L in the Nakdong River (median = 12.65), and 2.18 to 42.19 ng/L in the Seomjin River (median = 18.18). Most target compounds were detected at high frequency across the rivers, except PFDS and PFBA, which were not detected in water samples. Among the medaka fish samples, PFOS consistently showed the highest bioaccumulation across sites, with BAFs ranging from 2,871 L/kg in Yeosu to 37,335 L/kg in Dangjin, reflecting its strong bioaccumulative potential in aquatic organisms. Risk assessment using water quality criteria (WQC) showed that mean concentrations of PFOS (4.92 ng/L), PFBS (1.26 ng/L) and PFOA (19.62 ng/L) in water samples were below the criteria continuous concentration (CCC), criteria maximum concentration (CMC) and avian wildlife value (AWV), suggesting a low immediate risk to aquatic organisms. Hung Mai DucNga Nguyen ThiJung Hyeon JiCheon Hwi SuCho Hyeon Seo - Infection characteristics of Diplostomum spp. metacercariae (DsMc) were analyzed in fishes from 9 major water systems of Korea. A total of 15,879 fishes in 79 species were examined by the artificial digestion method for 8 years (2013-2020). DsMc were detected in 48 (60.8%) out of 79 fish species examined. The prevalence was higher in Pseudogobio esocinus (61.5%), Squalidus chankaensis (56.9%) and Zacco platypus (44.8%). The infection intensities were 22.1, 9.4, 7.5 and 6.7 per fish infected in Hemibarbus longirostris, P. esocinus, Z. platypus, and S. chankaensis respectively. The higher prevalence was shown in fishes from Yeongsan-gang (59.3%) and the middle reaches of Seomjin-gang (48.0%), and the higher infection intensities were revealed in fishes from above 2 regions and streams in east coast (83.1, 7.5, and 11.3 per fish infected). In index fish, Z. platypus, overall prevalence was 44.8% and the higher prevalence were revealed in the middle reaches of Seomjin-gang (79.6%), Geum-gang (79.2%) and Tamjin-gang (64.7%). The susceptibility index in index fish was highest in fish from the streams in east coast (13.64), and followed by fish from the middle reaches of Seomjin-gang (7.56) and Mangyeong-gang (4.75). Sohn Woon-MokNa Byoung-KukKim Jung-AKim Hee-Ju - Mitochondrial dysfunction is a pivotal pathogenic mechanism in colorectal cancer (CRC), which is further exacerbated by dysregulated liquid-liquid phase separation (LLPS). However, the mechanisms underlying mitochondrial dysfunction and LLPS, as well as their therapeutic targeting, remain unclear. Using Raddeanin A as a probe and through multiomics analysis, we identified apoptosis and caspase activation inhibitor (AVEN) as a potential therapeutic target. AVEN was significantly upregulated in CRC tissues and localized near mitochondria-endoplasmic reticulum contact sites (MERCS). Biophysical assays confirmed that AVEN undergoes LLPS via its intrinsically disordered regions (IDRs). Critically, we found that AVEN IDRs orchestrate mitochondrial homeostasis through dual LLPS-dependent mechanisms: inhibiting apoptosis and facilitating post-damage mitochondrial transfer. Furthermore, we identified sumatriptan (SUM) as a potent inhibitor of AVEN IDRs, which demonstrated promising therapeutic efficacy in mouse models and clinical cohorts. Our study highlights the critical role of AVEN in regulating mitochondrial homeostasis via LLPS and suggests that targeting AVEN represents a promising therapeutic strategy for CRC. - Source: PubMed
