Despite its significance, a comprehensive pan-cancer analysis of ZNF703 remains underexplored. In this study, we performed a systematic pan-cancer analysis to elucidate the mechanistic and functional roles of ZNF703 in tumorigenesis. Our findings reveal that elevated ZNF703 expression is significantly correlated with cancer progression, adverse clinical outcomes, and the enrichment of immune cells within the tumor microenvironment (TME), particularly cancer-associated fibroblasts (CAFs), CD8 T cells, and M2 macrophages, suggesting its pivotal role in modulating tumor immunity. Mechanistically, ZNF703 regulates tumor immunity by binding to promoter sequences, thereby suppressing the expression of CD274, ICAM1, and CXCL3, which may facilitate tumor immune escape. Additionally, we identified functional hub genes associated with ZNF703, including DDHD2, LSM1, and BAG4. Notably, ZNF703, DDHD2, LSM1, and BAG4 are co-localized within the amplicons at the chromosome 8p11-p12 region, indicating a potential cooperative role in driving cancer initiation and progression. Collectively, these findings underscore the essential roles of ZNF703 in cancer development, patient prognosis, and the regulation of anti-tumor immunity, highlighting its potential as a biomarker for cancer detection and as a novel immunotherapeutic target. - Source: PubMed

This study aimed to investigate the expression levels, diagnostic and prognostic significance and biological roles of Bcl-2-associated athanogene 4 (BAG4) in AML carcinogenesis. : Gene expression profiles were analyzed using publicly available datasets, particularly GSE9476 and TCGA, using tools such as GEO2R, GEPIA2, UALCAN and TIMER2.0. The immune infiltration correlation was examined using the GSCA platform, while the function of BAG4 at the single-cell level was analyzed via CancerSEA. Protein-protein and gene-gene interaction networks were constructed using STRING and GeneMANIA, and enrichment analyses were performed using GO, KEGG and DAVID. Expression validation was performed using RT-qPCR in HL-60 (AML) and HaCaT (normal) cells, and ROC curve analysis evaluated the diagnostic accuracy. : BAG4 was significantly overexpressed in AML tissues and cell lines compared with healthy controls. High BAG4 expression was associated with poor overall survival and strong diagnostic power (AUC = 0.944). BAG4 was positively associated with immune cell infiltration and negatively associated with CD4+/CD8+ T and NK cells. At the single-cell level, BAG4 was associated with proliferation, invasion, and DNA repair functions. Functional network analysis showed that BAG4 interacted with apoptosis and necroptosis-related genes such as BCL2, BAG3 and TNFRSF1A and was enriched in pathways such as NF-κB, TNF signaling and apoptosis. : BAG4 is overexpressed in AML and is associated with adverse clinical outcomes and immune modulation. It may play an important role in leukemogenesis by affecting apoptotic resistance and immune evasion. Akidan OsmanYaman SelçukOzer Yaman SerapMisir Sema - The depletion of freshwater resources constitutes a significant global issue, prompting the consideration of treated sewage wastewater (SWW) reuse as a viable alternative water source within contemporary water management strategies. The current study represents an innovative silver borate-based bioglass (BG) system for the treatment of municipal SWW. The melting-quenching method was used to prepare silver doped BG samples with different AgO concentrations. BAg1, BAg2, BAg3, and BAg4 BG samples were obtained from (65-X) BO-20NaO-10CaO-5PO-XAgO (where X = 0, 1, 2, 3, and 4 mol%). A major fecal coliform bacterium, Escherichia coli O157:H7 (E. coli O157:H7), was used as an indication of SWW purification. BG samples were characterized using FTIR, XRD, SEM, EDX, and DLS, respectively. BAg1 BG sample showed the strongest antibacterial activity against E. coli O157:H7, with a 27.0 ± 1.0 mm inhibition zone and 95.31% growth inhibition. Compared to other BG samples, the inhibition zone diameter was marginally lower than the amoxicillin positive control (28.33 ± 0.1 mm). Additionally, all BG samples inhibit the coliform bacterial community approximately 100%., whereas the original BG sample (BAg0) did not inhibit coliform bacteria. All BG samples had minimum inhibition concentrations (MICs) of 5 mg/mL and minimum bactericidal concentrations (MBCs) of 50 mg/mL, respectively, compared to the untreated bacterial culture (- Ve control). SEM micrographs showed that BAg1 sample treated E. coli O157:H7 cells deteriorated more than BAg0. In conclusion, silver-doped BG is a cost-effective, eco-friendly, and highly recommended technology for SWW purification from fecal coliform bacteria and therefore, can be used for reclamation and other non-drinking water. - Source: PubMed
