- Postmenopausal osteoporosis (PMOP) is a metabolic bone disorder driven by excessive osteoclast (OC) activity, but current therapies are limited by adverse effects. Trans-zeatin (TZ), a naturally occurring plant cytokinin with antioxidant properties, has not been investigated in bone metabolism. Here, we show that TZ suppresses receptor activator of nuclear factor-κB ligand (RANKL)-induced osteoclastogenesis and bone resorption in vitro and attenuates ovariectomy-induced trabecular bone loss in mice. Mechanistically, TZ activates nuclear factor erythroid 2-related factor 2 (Nrf2)-mediated antioxidant responses by upregulating heme oxygenase-1 (HO-1) and catalase while suppressing NADPH oxidase 4 (NOX4) expression, thereby reducing reactive oxygen species (ROS) accumulation. This redox modulation subsequently dampens protein kinase B (AKT) signaling, downregulates the c-Fos/nuclear factor of activated T-cells cytoplasmic 1 (NFATc1) transcriptional program, and decreases the expression of OC-specific effector genes. Notably, Nrf2 knockdown partially reverses the inhibitory effects of TZ, whereas concurrent AKT knockdown attenuates this rescue effect, suggesting that AKT is an important downstream signaling component associated with Nrf2-mediated redox regulation. Collectively, these findings identify TZ as a promising natural candidate for PMOP intervention through coordinated regulation of redox homeostasis and AKT-dependent osteoclastogenic signaling. - Source: PubMed

Here, we present a rapid and efficient method for generating multiplex gene knockout cells from THP-1 using a simple co-transfection strategy based on the CRISPR-Cas9 system. In this method, THP-1 cells were transfected with a mixture of pX330-based sgRNA plasmids and a small amount of puromycin-resistance plasmid by electroporation, followed by transient selection with high-dose puromycin. This simple method resulted in efficient simultaneous knockout of multiple genes. Among them, fructooligosaccharides and galactooligosaccharides are especially important for their positive effects on human health. Functional foods with enhanced nutritional and medicinal value have been made possible by the recent use of prebiotics in food fermentation and bioprocessing. This review summarizes recent developments in prebiotic production and application, with emphasis on biotechnological approaches such as microbial fermentation, enzymatic synthesis, and strain improvement techniques, based on literature retrieved from PubMed, Scopus, and Web of Science up to July 2025. Advances in fermentation technologies, including solid-state fermentation and co-culture systems, are also discussed for optimizing production processes. In addition, the review highlights the health-promoting effects of prebiotics, including modulation of gut microbiota, immunological regulation, and involvement in the gut-brain axis. The experimental results demonstrated that topical application of rhCol III gel to dorsal skin wounds in rats significantly increased the wound healing rate. Further analysis revealed that rhCol III gel improved the extracellular matrix (ECM) environment and mediated an increase in the expression of various integrins. Through interaction with integrins, rhCol III gel transmitted extracellular signals into the cell the ECM-receptor signaling pathway, positively regulating the PI3K-Akt signaling pathway. This led to upregulated expression of genes such as Fos, Nos3, and Tnc, ultimately inducing the regeneration of extracellular matrix components, including collagen type I and III, elastin, and fibronectin, thereby achieving ECM remodeling and skin wound repair. Hematoxylin and eosin (HE) staining, as well as analyses of routine blood tests, serum biochemical analysis, and coagulation function tests, showed no significant differences between the experimental and control groups. Therefore, rhCol III gel exhibits high efficacy in tissue repair and favorable safety, demonstrating a promising safe and effective treatment for skin injury repair. - Source: PubMed
