Clinical, histopathologic, immunohistochemical, next-generation sequencing (NGS), and follow-up data were collected, and related studies were reviewed.ResultsAll tumors were detected by imaging studies. Ultrasound revealed well-circumscribed echogenic renal masses. Tumor tissue was separated by a fibrovascular network into nests, sheets, or acini, with varying amounts of melanin pigmentation. The tumor cells were epithelioid in shape, with clear or eosinophilic cytoplasm and prominent nucleoli. They were positive for TFE3 and HMB45 but negative for Melan-A, PAX8, CA9, CD10, keratin, vimentin, S100, SMA, and desmin. Notably, the tumor from the 28-year-old female patient demonstrated marked atypia, tumor necrosis, dystrophic calcification, and atypical mitotic figures. The fusion gene was detected in all 3 patients by NGS. Missense mutations in the , , , and (tuberous sclerosis complex, TSC) gene were also detected. All patients survived without recurrence or metastasis.ConclusionsRenal TFE3-rearranged PEComa display unique clinicopathologic features, usually with insidious onset and potential for recurrence, metastasis, or death, and may represent an independent tumor subtype. Differential diagnoses include conventional perivascular epithelioid cell tumors and Xp11 translocation renal cell carcinoma. Zhou ZidiLei TingShi YongqiangWang HuiPeng YanLi QingChen Tongbing - This study aimed to investigate the regulatory effects of replacing soybean meal with varying proportions of fermented cottonseed meal in the diet on protein metabolism in the pectoral and leg muscles of white-feather broilers. The experiment was divided into two phases: early-growth (1-21 days) and late-growth (22-42 days) phases. The control group was fed a basal diet without fermented cottonseed meal (0%), whereas the experimental groups were fed soybean meal replaced with 3, 6, and 9% fermented cottonseed meal. The effect on protein metabolism was evaluated by measuring the expression of key genes in the IGF1/mTOR, AMPK, and UPP pathways in muscle tissue. During days 1-21, experimental groups showed significantly higher IGF-1 and mTOR mRNA expression in leg muscles compared to controls, while FoxO3 expression in pectoral muscles was reduced. The 3 and 6% groups had elevated IGF-1 and PI3K, and the 6% group showed higher AKT and mTOR in pectoral muscles and PI3K in leg muscles. TSC2 expression was suppressed in the 3 and 6% groups, while Atrogin-1 and MuRF1 were lowest in the 6% group. During days 22-42, the 3 and 9% groups had increased IGF-1 and mTOR in pectoral muscles, whereas the 6% group showed higher AKT and PI3K in leg muscles. FoxO3 was significantly downregulated in the 9% group. In summary, fermented cottonseed meal can regulate protein metabolism in the pectoral and leg muscles of Cobb broilers by enhancing the expression of genes related to the IGF-1/mTOR pathway and suppressing the transcription of certain key genes in the UPP, thereby promoting protein deposition, with an optimal supplementation level of 6%. - Source: PubMed

We introduce a single-episode differential signaled active avoidance (DSAA) paradigm that temporally dissociates acquisition, consolidation, and retrieval of instrumental avoidance memory. Interleaving a behaviorally noncontingent neutral cue (CS-) with a threat-predictive, behaviorally contingent cue (CS+) enhanced long-term memory without altering acquisition, demonstrating that differential contingency structure selectively reinforces memory consolidation rather than influencing learning performance. Naïve animals inferred contingencies within a single episode, and discrimination achieved during training predicted retrieval precision. However, discrimination operated within defined boundary conditions: overtraining or elevated threat intensity destabilized cue specificity and promoted persistent avoidance generalization. Under high-threat conditions, freezing and shuttling co-emerged as complementary defensive responses, indicating a shift from precise cue-based encoding to a generalized defensive state. Remote retrieval recruited oxytocin receptor-expressing cells in the medial prefrontal cortex and activated mTORC1-dependent translational signaling, implicating protein synthesis in maintenance of discriminative avoidance memory. In a Tuberous Sclerosis Complex model with Tsc2 haploinsufficiency in oxytocin-responsive cells, males displayed intact acquisition but generalized avoidance at both recent and remote time points, a deficit not rescued by additional training. These findings identify oxytocin-modulated translational control as a molecular gate stabilizing threat-safety discrimination and show that disruption of this axis - by excessive threat or reduced Tsc2 gene dosage - biases memory toward pathological generalization, providing a mechanistic framework for safety-learning deficits in neurodevelopmental and anxiety-related disorders. - Source: PubMed
