This report describes a study performed in 66 healthy volunteers to assess the persistent antimicrobial efficacy of a commercially available skin antiseptic composed of 2% CHG and 70% isopropyl alcohol (IPA). The solution was applied to the abdominal and inguinal (groin) regions and was allowed to dry before being assessed for immediate effect and covered with dressings. The antimicrobial persistence was examined at different time points over ten days post-application and the reduction in skin bacterial levels (CFU/cm) from baseline was assessed. The mean Log reduction in bacteria on the abdomen was 2.75 at 10 minutes, 2.03 at 7 days, and 1.53 at 10 days post-application. The mean Log reduction in skin bacteria on the groin was 3.59 at 10 minutes, 2.80 at 7 days, and 2.42 at 10 days post-application. The results showed that the antimicrobial effect of this skin prep can be sustained for 10 days post-application when used on healthy volunteers. Further studies are needed to determine the antimicrobial persistence and effect on SSIs when this antiseptic agent is used preoperatively. - Source: PubMed

However, its enzymatic synthesis from sucrose and lactose is often limited by glucose accumulation, which shifts the reaction equilibrium toward hydrolysis. In this study, we developed an efficient preparative production system for lactosucrose by integrating -derived levansucrase with an invertase-deficient ATCC 56741. Unlike conventional wine yeasts, the selected strain preferentially reduced glucose while minimizing sucrose and lactosucrose degradation under the tested conditions, thereby maintaining favorable conditions for transfructosylation. The integrated system was optimized by evaluating various relative proportions of enzyme activity and yeast biomass standardized by cell density. Under the optimized yeast-dominant condition (activity-to-biomass ratio of 1:3), a maximum lactosucrose concentration of 128.2 g/L was achieved. Furthermore, the biological removal of glucose may simplify downstream processing by reducing byproduct interference. Unlike glucose oxidase-based systems, which generate gluconic acid and require pH control, the present approach utilizes biological glucose removal without introducing additional acidic byproducts. Han Woo-CheulLee Je-HyukJang Ki-Hyo - We developed a pre-exposure prophylaxis (PrEP) shared decision-making tool to support people who could benefit from PrEP in reaching their prevention goals and preferences when considering two effective PrEP modalities, oral or long-acting injectable (LAI) PrEP, in partnership with their healthcare providers (HCPs). This 2-phase study included formative research to develop a PrEP shared decision-making tool prototype followed by a pilot study to determine prototype acceptability. For the formative phase, we conducted semi-structured qualitative in-depth interviews with a diverse sample of people who could benefit from PrEP (n = 41) and HCPs (n = 20) in two community-based sites in Washington, DC, prior to and shortly after LAI PrEP became available in December 2021. Insights from in-depth interviews informed development of a PrEP shared decision-making tool prototype in both English and Spanish. This prototype was piloted in one-on-one mock clinical encounters with 37 people who could benefit from PrEP and 11 HCPs, in the two DC sites and at a university-affiliated clinic in Chapel Hill, North Carolina. After these encounters, people who could benefit from PrEP and HCPs were interviewed independently about acceptability of the prototype's language, visuals, content and flow as well as the usability, utility, and feasibility of the prototype to support informed shared decision-making. In the formative phase, people who could benefit from PrEP had low awareness of and high interest in LAI PrEP. They and HCPs agreed that side effects and efficacy should be a primary focus of a PrEP shared decision-making tool. Participants in the mock clinical encounters found that the prototype filled information gaps, supported shared decision-making regardless of an individual's experience with PrEP, and systematized, streamlined and normalized conversations around PrEP choice. Testing the effectiveness of the prototype in a larger-scale evaluation is highly warranted as the field seeks to address ongoing inequities in PrEP uptake. - Source: PubMed
