- Bone metabolism is regulated by immune cells that maintain skeletal homeostasis through cytokine secretion and cell interactions. Disruption of this regulation-by inflammation, autoimmunity, or aging-drives pathological bone diseases including osteoporosis and rheumatoid arthritis. Single-cell technologies and exosome biology have revealed previously unrecognized immune subsets and regulatory layers within the bone microenvironment. - Source: PubMed

Jin ZhuoruLiu LishengWang HeranWei YufeiZhou ZimoLuo XiaohuiLiu Da - Macrophage-associated responses at sites of nerve injury are involved in the initiation and persistence of neuropathic pain (NP). Immune semaphorins (SEMAs), including SEMA3A, SEMA3E, SEMA4A, SEMA4D, and SEMA7A, regulate macrophage migration and activation, but their roles in NP remain unclear. This study aimed to identify immune SEMAs associated with NP by analyzing their serum levels and expression in sensory nerve tissues of patients with NP, and to evaluate the potential effects of SEMA-targeted intervention in a mouse model. - Source: PubMed

Okutucu MuratArpa Medeni - The nervous system plays a crucial role in regulating bone remodeling, with both sensory and autonomic nerves distributed within bone tissue. In the mandible, the inferior alveolar nerve (IAN) and sympathetic nerves provide the primary innervation. Numerous studies have demonstrated their essential functions in mandibular development and bone metabolism. Focusing on mandibular innervation, we synthesize current evidence to demonstrate the modulatory effects of sensory and sympathetic nerves on bone growth and regeneration, and further elucidate the underlying mechanisms. Notably, we discuss multiple bioactive factors (CGRP, SP, VIP, NE, NPY, Sema3A, NGF) for their roles in mediating neural regulation of bone. Additionally, neuro-bone crosstalk in the mandible is examined in an integrated way. This review offers a deeper understanding of mandibular homeostasis modulated by nerves and thus seeks to facilitate future clinical translation. - Source: PubMed
