- This study aimed to explore the molecular alterations in airway epithelial cells in children with wheezing infected by respiratory syncytial virus (WheezeRSV). This study extracted the single-cell sequencing data of two control and wheezeRSV samples from GSE286262 dataset. The monocle2 was performed for analyzing evolution process of basal cells along disease progression. Kyoto encyclopedia of genes and genomes and gene ontology were carried out to identify the function enrichment of differentially expressed genes (DEGs) between WheezeRSV and control groups. Additionally, human nasal epithelial cells (HNECs) were harvested, and quantitative real-time polymerase chain reaction (qRT-PCR) was applied to detect the expression levels of key genes. Six cell types including ciliated cells, KRT5 + basal cells, TP63 + basal cells, KRT80 + epithelial cells, goblet cells, and club cells. The differentiation trajectory analysis on basal cells indicated two distinct branches including ciliary assembly and keratinization. Then, function analysis indicated that ciliated cells were involved in leukocyte transendothelial migration, cellular senescence and angiogenesis. Goblet cells were associated with IL-17 signaling pathway, apoptosis, and ferroptosis, and club cells were enriched in the apoptotic process and innate immune response. The qRT-PCR results revealed that the mRNA expression levels of , , , , , and were markedly upregulated in the WheezeRSV group ( < 0.01). Basal cells differentiated into keratinocytes and ciliated cells as the disease progresses, thereby enhancing defense and repairing the epithelial barrier. Meanwhile, wheezeRSV enhanced the activity of apoptosis and inflammatory response of ciliated cells, goblet cells, and club cells. - Source: PubMed

Wei ZhengboWang LifangDing XingHaiWang ChengzhongLi Sheng - Infertility is a major global health concern, with male factors accounting for nearly half of all cases. While tektin proteins are known to stabilize flagellar microtubules, comprehensive validations of the TEKT gene family across diverse human infertility phenotypes remain limited. This study aimed to evaluate the expression and regulatory genetics of the TEKT family, particularly TEKT2, to determine its specific role in human spermatogenesis and sperm morphological disorders. - Source: PubMed

Kakavand KianoushHosseini Seyedeh HaniehHeydari RahelehHaratian KavehSabbaghian MarjanMeybodi Anahita Mohseni - Induction of spawning in catfish is well demonstrated in females while stimulation of spermiation in males seems difficult. This has led to least understanding of gene profile changes during testicular maturation. Expression of the factors after hormonal induction that control reproductive processes has become an intense research area in fish endocrinology. In view of this, de novo assemblies of testicular transcriptomes of the Asian catfish, Clarias batrachus and the African catfish, C. gariepinus, were performed to identify genes/factors involved in testicular maturation. For testicular induction, human chorionic gonadotropin (hCG) was administered in vivo to both the catfish species during active phase of reproduction. 1,68,071 and 1,26,232 assembled transcripts were obtained from C. batrachus and C. gariepinus testicular transcriptomes and were annotated using different databases. Further, in silico analysis suggested the presence of several transcripts that were involved in steroidogenesis and male reproduction. Comparison of transcriptomes of both species revealed the presence of certain unique genes related to reproduction differentially. Expression profiles after hCG induction in testis showed higher expression of certain steroidogenesis related genes such as star, cyp11b1, cyp17a and cyp21a in both the species. Further, expression levels of crucial factors related to testis, such as dmrt1/2/3, were also found to be significantly higher after gonadotropin induction. However, amh, tgfα and dmrt4/5 expression levels were significantly low. Factors related to male reproduction i.e., tekt1, tekt2, sox9, spag1, spata2 and spata7 were found to be differentially upregulated in hCG treated testicular tissues of both the species. Histology analysis indicated that the gonadotropin induction either short or long term is a better strategy to highlight expression profile changes during testicular maturation in teleost. - Source: PubMed

Kar SonikaTenugu SwathiPranoty AkankshaAnusha NarlagiriUdit Uday KumarSangem PrabhakerSenthilkumaran Balasubramanian - Vertebrate left-right symmetry breaking is preceded by formation of left-right organizer. In Amphibian, this structure is formed by gastrocoel roof plate, which emerges from superficial suprablastoporal cells. GRP is subdivided into medial area, which generates leftward flow by rotating monocilia and lateral Nodal1 expressing areas, which are involved in sensing of the flow. After successful symmetry breaking, medial cells are incorporated into a deep layer where they contribute to the axial mesoderm, while lateral domains join somitic mesoderm. - Source: PubMed

Petri NataliaVetrova AlexandraTsikolia NikolozKremnyov Stanislav - Spermatogenesis is a complex process related to male infertility. Till now, the critical genes and specific mechanisms have not been elucidated clearly. Our objective was to determine the hub genes that play a crucial role in spermatogenesis by analyzing the differentially expressed genes (DEGs) present in non-obstructive azoospermia (NOA) compared to OA and normal samples using bioinformatics analysis. Four datasets, namely GSE45885, GSE45887, GSE9210 and GSE145467 were used. Functional enrichment analyses were performed on the DEGs. Hub genes were identified based on protein-protein interactions between DEGs. The expression of the hub genes was further examined in the testicular germ cell tumors from the TCGA by the GEPIA and validated by qRT-PCR in the testes of lipopolysaccharide-induced acute orchitis mice with impaired spermatogenesis. A total of 203 DEGs including 34 up-regulated and 169 down-regulated were identified. Functional enrichment analysis showed DEGs were mainly involved in microtubule motility, the process of cell growth and protein transport. PRM2, TEKT2, FSCN3, UBQLN3, SPATS1 and GTSF1L were identified and validated as hub genes for spermatogenesis. Three of them (PRM2, FSCN3 and TEKT2) were significantly down-regulated in the testicular germ cell tumors and their methylation levels were associated with the pathogenesis. In summary, the hub genes identified may be related to spermatogenesis and may act as potential therapeutic targets for NOA and testicular germ cell tumors. - Source: PubMed
