Despite the pivotal role of the hypothalamic-pituitary-gonadal (HPG) axis in avian reproduction, the underlying molecular mechanisms that govern sexual maturity and spermatogenesis in roosters are not well elucidated. This study aimed to explore gene expression changes in the HPG axis using Wenchang cockerels.2. Phenotypic results showed significant testicular growth from 18 to 20 weeks of age. Transcriptome analyses identified 2,154 differentially expressed genes (DEG) in testes, which was markedly higher than those identified in hypothalamus (52 DEG) and pituitary (27 DEG).3. Functional enrichment analysis showed neuroactive ligand-receptor interactions and hormone transport pathways in the hypothalamus and pituitary and ciliary motility and spermatogenesis pathways in the testes.4. Protein-protein interaction (PPI) networks analysis pinpointed key hub genes, such as , , , , , and , associated with spermatogenesis and sperm motility.5. Liu YAzim SLi JZhang BZhang ZLuo XSu CLiu XFeng Y - This study aimed to explore the molecular alterations in airway epithelial cells in children with wheezing infected by respiratory syncytial virus (WheezeRSV). This study extracted the single-cell sequencing data of two control and wheezeRSV samples from GSE286262 dataset. The monocle2 was performed for analyzing evolution process of basal cells along disease progression. Kyoto encyclopedia of genes and genomes and gene ontology were carried out to identify the function enrichment of differentially expressed genes (DEGs) between WheezeRSV and control groups. Additionally, human nasal epithelial cells (HNECs) were harvested, and quantitative real-time polymerase chain reaction (qRT-PCR) was applied to detect the expression levels of key genes. Six cell types including ciliated cells, KRT5 + basal cells, TP63 + basal cells, KRT80 + epithelial cells, goblet cells, and club cells. The differentiation trajectory analysis on basal cells indicated two distinct branches including ciliary assembly and keratinization. Then, function analysis indicated that ciliated cells were involved in leukocyte transendothelial migration, cellular senescence and angiogenesis. Goblet cells were associated with IL-17 signaling pathway, apoptosis, and ferroptosis, and club cells were enriched in the apoptotic process and innate immune response. The qRT-PCR results revealed that the mRNA expression levels of , , , , , and were markedly upregulated in the WheezeRSV group ( < 0.01). Basal cells differentiated into keratinocytes and ciliated cells as the disease progresses, thereby enhancing defense and repairing the epithelial barrier. This has led to least understanding of gene profile changes during testicular maturation. Expression of the factors after hormonal induction that control reproductive processes has become an intense research area in fish endocrinology. In view of this, de novo assemblies of testicular transcriptomes of the Asian catfish, Clarias batrachus and the African catfish, C. gariepinus, were performed to identify genes/factors involved in testicular maturation. For testicular induction, human chorionic gonadotropin (hCG) was administered in vivo to both the catfish species during active phase of reproduction. 1,68,071 and 1,26,232 assembled transcripts were obtained from C. batrachus and C. gariepinus testicular transcriptomes and were annotated using different databases. Further, in silico analysis suggested the presence of several transcripts that were involved in steroidogenesis and male reproduction. Comparison of transcriptomes of both species revealed the presence of certain unique genes related to reproduction differentially. Expression profiles after hCG induction in testis showed higher expression of certain steroidogenesis related genes such as star, cyp11b1, cyp17a and cyp21a in both the species. Further, expression levels of crucial factors related to testis, such as dmrt1/2/3, were also found to be significantly higher after gonadotropin induction. However, amh, tgfα and dmrt4/5 expression levels were significantly low. Factors related to male reproduction i.e., tekt1, tekt2, sox9, spag1, spata2 and spata7 were found to be differentially upregulated in hCG treated testicular tissues of both the species. Histology analysis indicated that the gonadotropin induction either short or long term is a better strategy to highlight expression profile changes during testicular maturation in teleost. - Source: PubMed
