- Lactation traits are important indicators for evaluating the production performance of dairy cows and the economic efficiency of dairy production. Identifying potential regulatory genes is essential for elucidating the molecular mechanisms underlying lactation and facilitating molecular breeding. This study aimed to identify candidate genes and regulatory pathways potentially associated with lactation traits in dairy cows by integrating transcriptome expression profiles of primary bovine mammary epithelial cells (BMECs) from eight Holstein cows with lactation phenotypic data. A gene co-expression network was constructed using weighted gene co-expression network analysis (WGCNA). Co-expression modules associated with lactation traits were identified, and candidate genes were further screened by integrating gene significance, module membership, functional enrichment analysis, random forest analysis, gene-phenotype association analysis, single-gene gene set enrichment analysis (GSEA), ROC curve analysis, and tissue expression profiling. Four co-expression modules, namely MEdarkturquoise, MEsteelblue, MEbrown, and MEskyblue3, were significantly associated with lactation traits ( < 0.05). Genes in these modules were mainly enriched in biological processes and pathways related to ribosome biogenesis, protein translation, the cell cycle, oxidative phosphorylation, and the PI3K-Akt signaling pathway, which may be involved in lactation regulation. Through multi-strategy cross-screening, four candidate genes were ultimately identified. 2 was associated with daily milk yield (DYM), A with total milk solids (TMS), and 25 and 2 with milk fat percentage (MFP) and fat-to-protein percentage ratio (FPP). Reverse transcription quantitative real-time PCR (RT-qPCR) validation showed that 2 and were relatively highly expressed in mammary tissue, suggesting that they may be involved in lactation-related biological processes in dairy cows. These findings provide potential candidate regulators and theoretical support for further studies on the molecular mechanisms of lactation traits; however, their functional roles require further validation through in vitro and in vivo experiments. - Source: PubMed

However, a multi-tissue transcriptomic atlas for the key dairy river buffalo breeds, Murrah and Nili-Ravi, has not yet been established, and the lack of stable reference genes has hindered in-depth studies of their biological functions and the molecular mechanisms underlying key economic traits such as lactation. We established a multi-tissue gene expression atlas across 20 tissues and identified 717 housekeeping genes (HKGs), and and were further shown to be stable candidate reference genes under the conditions tested. We found 8368 tissue-specific genes (TSGs), predominantly enriched in the reproductive system. Exploratory analysis of mammary tissue (dry-period vs. public lactating samples, confounded by batch effects) revealed mammary-enriched hub genes including ; these findings are preliminary and require validation. Dynamic analysis across lactation stages (early, peak, mid-, late) identified candidate genes including and . Phenotypic data showed strong negative correlations between milk yield and protein/fat content, and a positive correlation with lactose content. However, causal or regulatory roles were not inferred due to lack of paired individual-level data. Cross-dataset comparisons are descriptive only, and are not key conclusions. Employing an integrated multiomics framework, this study combined bidirectional Mendelian randomization (MR), network toxicology, machine learning, molecular simulations, and ovariectomized rat models to elucidate causal relationships between EDCs and osteoporosis, and to identify the molecular underpinnings of these relationships. MR analyses leveraging European GWAS data identified methylparaben (MP; OR = 0.973, < 0.001) and monomethyl phthalate (MMP; OR = 0.984, p = 0.006) as causal agents reducing bone mineral density (BMD), validated across two independent cohorts. Network toxicology revealed CYP3A4 as a shared target for both EDCs, with HSPA5-driven endoplasmic reticulum (ER) stress implicated in MP-induced bone loss and CRP-mediated inflammation central to MMP pathology. Molecular dynamics simulations confirmed stable binding of MP/MMP with hub targets. Crucially, ribosomal genes (RPL9/RPL37A/RPS19) altered by MP were mechanistically linked to ER stress-induced osteotoxicity rather than direct EDC binding. In vivo validation demonstrated that MP and MMP exposure in OVX rats significantly exacerbated trabecular degradation, suppressed osteogenic markers, and elevated osteoclastic activity. This work establishes CYP3A4 as a high-value therapeutic target for countering EDC-induced osteoporosis and resolves longstanding controversies regarding the osteotoxic mechanisms of parabens and phthalates. - Source: PubMed
