- Breast cancer (BC) is a major global health concern, ranking among the most common neoplasms and representing one of the leading causes of cancer-related deaths worldwide. Early recognition and classification of BC subtypes are crucial for improving patient outcomes. Therefore, identifying novel biomarkers with diagnostic and prognostic significance is of great importance. The Wnt signaling pathway plays a significant role in BC by influencing various cell cycle regulation processes and stem cell renewal. This study aims to identify novel Wnt-associated biomarker panels for BC patients, composed of multiple molecular factors. A series of bioinformatical analyses have been employed, including weighted gene co-expression network analysis, differential expression analysis, Kaplan-Meier survival analysis, logistic regression model evaluation, and receiver operating characteristic construction. Thus, this study revealed potential diagnostic and prognostic signatures based on comprehensive analyses of BC patient data sourced from The Cancer Genome Atlas database. Consequently, four gene signatures were constructed: two differentiate ER+ from ER-BC: and for overall survival (OS); and for disease free survival (DFS), while the other two effectively distinguish tumor from normal samples: for OS; and for DFS. - Source: PubMed

Li JingJiang FengeWang CongcongSun PingSong LeiLiu Jiannan - This study aimed to identify genomic regions associated with prolificacy in Santa Inês sheep raised in tropical conditions. The prolificacy of the dam was defined as single (only one lamb born per ewe per lambing) or multiple (more than one animal born per ewe per lambing). After quality control of phenotypic data, 1584 lambing records of 715 females occurred between the years 2000 and 2018 were used. The animals were genotyped with the OvineSNP50 BeadChip panel (Illumina Inc.). After quality control of genomic data, information of 46,714 SNPs and 388 samples and females was used for the subsequent analyses. The single-step GWAS (ssGWAS) methodology was used to estimate the effects of genetic markers and their association with the prolificacy. A total of 21 windows of 10 adjacent SNPs that explained at least 0.5% of the additive genetic variance for prolificacy were identified. In such regions, genes associated with different reproductive functions in the female were found: CACNA1E, NTRK1, PLCH1, SMAD3, CENPF, TOPBP1, IL33, DRD2, MID1, HCCS, and ARHGAP6. Some candidate regions related to prolificacy harbor genes that were not previously described and genes without known functions. These results can help to identify genes associated with prolificacy and could be used in genomic reproductive studies on prolificacy, as well as in the selection of the most prolific ewes in the population. - Source: PubMed

Several brain PLC subtypes have been extensively studied, implicating them in psychiatric disorders such as depression, epilepsy and schizophrenia. However, the role of the recently identified PLCη remains largely unknown. We found that PLCη1 is prominently expressed in lateral habenula (LHb) astrocytes. Here, to investigate its physiological role, we generated astrocyte-specific PLCη1 conditional knockout (cKO) mice (Plch1; Aldh1l1-Cre). In these cKO mice, we observed a reduction in cellular morphological complexity metrics, such as total process length, as well as a decrease in the passive membrane conductance of LHb astrocytes. Additionally, neuronal function was impacted by the cKO, as the synaptic efficacy and firing rates of LHb neurons increased, while extrasynaptic long-term depression was impaired. Both tonic α-amino-3-hydroxy-5-methyl-4-isoxazolepdlropionic acid receptor/N-methyl-D-aspartate receptor (AMPAR/NMDAR) currents and extracellular glutamate levels were reduced. Interestingly, chemogenetic activation of astrocytes restored the reduced tonic AMPAR/NMDAR currents in cKO mice. Furthermore, LHb astrocyte-specific deletion of PLCη1 via AAV-GFAP-Cre injection induced depressive-like behaviors in mice, which were reversed by chemogenetic activation of LHb astrocytes. Finally, we found that restraint stress exposure decreased Plch1 mRNA expression in the LHb. These findings suggest that PLCη1 could be a potential therapeutic target for depression and highlight the critical role of astrocytes in the etiology of neuropsychiatric disorders. - Source: PubMed
