- Retinoids (i.e., vitamin A and metabolites) have biological importance for functions involved in growth and development, immunity, and reproduction. Previous studies involving retinoids have demonstrated positive associations with increased luteal progesterone (P4) concentrations, and similarly, progesterone has been implicated in the regulation of uterine secretion of retinol-binding protein (RBP), warranting further investigation. Therefore, the aim of this study was to identify the expression of various enzymes and binding proteins involved in vitamin A metabolism in corpora lutea (CL) and endometria during day 15 of the estrous cycle and throughout various points of gestation in gilts. Sexually mature, crossbred gilts were ovariohysterectomized on days 11, 15, 60, or 90 of pregnancy, or day 15 of the estrous cycle (cyclic day 15). Plasma was collected for P4 and vitamin A analysis. Corpora luteal and endometrial tissues were analyzed for mRNAs associated with vitamin A metabolism. There were no differences in CL count, P4 concentrations, or vitamin A concentrations by cyclic or gestational day (P > 0.05). Corpora lutea expression of aldehyde dehydrogenase 1 family member A1 (ALDH1A1) and RBP4 mRNAs were greater on day 15 of pregnancy compared cyclic day 15 gilts (P < 0.05). Moreover, corpora lutea mRNA expression of ALDH1A1 (P = 0.0003), RBP2 (P = 0.02), and alcohol dehydrogenase 1C (ADH1C, P = 0.07) were greater on day 11 of pregnancy. Corpora lutea expression of lecithin retinol acyltransferase (LRAT) mRNA were not different (P > 0.05). Endometrial mRNA expression of ALDH1A1 was greater on cyclic day 15 compared to day 15 of pregnancy (P = 0.002). Endometrial mRNA expression of ALDH1A1 and LRAT were greater on days 60 and 90 of pregnancy compared to days 11 and 15 of pregnancy (P ≤ 0.0001). Endometrial expression of RBP4 mRNA was greater on days 11 and 60 compared to day 90, and greater on day 60 compared to day 15 (P = 0.01). Endometrial expression of ADH1C and RBP2 mRNAs were not different (P > 0.05). These results suggest that genes encoding enzymes involved in vitamin A metabolism (ALDH1A1, ADH1C, RBP2, and RBP4) are elevated in the CL during the period of early gestation when the anti-luteolytic mechanism of maternal recognition of pregnancy is occurring. Stuehr Matthew DHickman Kyle JWiegert Jeffrey GPoole Rebecca K - Per- and polyfluoroalkyl substances (PFAS) can reach remote environments mainly through atmospheric transport and deposition, whereas methylmercury (MeHg) is formed primarily through in-situ microbial methylation of inorganic mercury (Hg). However, their occurrence in high-altitude environments is still poorly understood. This study provides the first assessment of the vertical distribution, source apportionment, and risk implications of PFAS and MeHg in peat cores from two alpine glaciated lakes in northern, Pakistan. Total ∑PFAS concentrations ranged from 3.49 to 221.56 ng/g, while MeHg concentrations ranged from 0.27 to 11.24 ng/g, with pronounced spatial variability. Naltar lake had higher PFAS concentrations, whereas Sheosar Lake showed higher MeHg concentrations. Long range atmospheric transport (LRAT) and glacial melt were identified as major PFAS sources, while MeHg was primarily associated with in-situ methylation of Hg. Risk assessment identified Perfluoroalkyl Carboxylic acids (PFCAs) and Perfluoroalkyl Sulfonic acids (PFSAs) as the compound classes with the highest risk quotients at Naltar lake, while MeHg was the main concern at Sheosar lake. Alpine peatlands serve as critical contaminant reservoirs and may act as secondary sources to downstream aquatic ecosystems amid ongoing glacial retreat. Pang SongJiang JunJiang Rui - Persistent organic pollutants (POPs), such as DDT and PCBs, accumulate in glaciers through long-range atmospheric transport (LRAT) and are re-released during melting. However, the potential for current-use pesticides (CUPs) to exhibit similar behaviour remains less understood. In this study, we employed (i) time-integrative passive sampling of meltwater streams across seven consecutive intervals (July-October 2024), (ii) spatially resolved surface sediments, and (iii) a dated sediment core (covering the period from ∼1970 to present), to investigate the occurrence and re-emission processes of CUPs in Alpine aquatic environments, alongside selected POPs as control substances. By comparing a glaciated catchment with a nearby catchment with minimal glacier cover in the Bernese Alps, Switzerland, 13 CUPs and their transformation products (TPs) were detected in the meltwater creeks. Several compounds were identified for the first time in this area, including fungicides (fluopicolide, and metalaxyl), herbicides (clomazone, and propyzamide), and transformation products (desethylterbuthylazine, and terbuthylazine-2-hydroxy). Time-weighted average water concentrations ranged from 2 to 860 pg/L, with peak concentrations occurring in early July and decreasing throughout the melt season, pinpointing specific compounds in each catchment. In addition, the sediment core reconstructions revealed that ΣDDT and ΣPCBs in sediments followed the anticipated trends of the mid-20th century but showed a significant increase starting in the 2000s, coinciding with accelerated glacier retreat. Surface sediments suggest a preferential association with silt and clay fractions, and projections of sediment-bound contaminants indicate potential inputs of up to 3.6 g ΣDDT, 2.2 g ΣPCBs, and 2.9 g CUPs over the next decade. Overall, our findings enhance the understanding of how catchment development influences post-depositional processes and highlight the role of snow and glacier melt as a persistent, continuous secondary source of contaminants to otherwise pristine ecosystems. - Source: PubMed
