- Vitamin D (VD) is a pleiotropic hormone with diverse physiological functions, but its role during early embryonic development in fish remains poorly understood. This study investigated the effects of aqueous VD exposure on embryonic survival, hatching, and the expression of the kiss1, kiss2, gnrh3, foxr1, and hsp70 genes in common carp (Cyprinus carpio L.). We also examined the hypothesis that common carp embryo is a site of VD non-genomic action and its metabolism, evaluated by measuring 25-hydroxyvitamin D (25OHD) levels and confirmed by immunolocalization of VDR, PDIA3 as well as CYP24A1 and CYP27B1. The eggs (obtained from 6 females) were fertilized and incubated in water containing VD at concentrations: 0, 1, 10, 50, and 100 ng/mL. During early embryonic development (24, 48, 72 h post-fertilization - hpf), VD did not reveal developmental abnormalities and mortality in common carp but increased larvae hatching rate at the highest tested concentration. Furthermore, early life exposure to VD has shown no alteration in kiss1, kiss2, gnrh3, foxr1, hsp70 gene expression in newly hatched larvae. Crucially, VD exposure altered embryonic 25OHD concentrations. This finding, alongside the immunohistochemical localization of VDR, PDIA3, CYP27B1 and CYP24A1 in the tissue of common carp embryo, indicates the possible genomic and non-genomic action of VD as well as its active metabolism (synthesis and inactivation) at early life stages of this species. By examining the rhythmic changes in these parameters under long (16L: 8D) and short (8L: 16D) photoperiods, we aim to elucidate the role of melatonin and photoperiod in regulating seasonal reproductive physiology. One hundred and ninety-two male Syrian hamsters were equally divided into four groups ( = 48/group). Two groups were pinealectomized (pinx) and the other two groups were sham-operated (control). Treatment and control groups were then exposed to either a long photoperiod (LP) of 16 hours light/8 hours dark or a short photoperiod (SP) of 8 hours light/16 hours dark for 30 days and then sampled at specific temporal intervals (ZT0, ZT4, ZT8, ZT12, ZT16, and ZT20) at which hypothalamic tissues and serum samples were collected. Kiss1 mRNA expression and protein levels were measured using qPCR and Western blotting, while testosterone and melatonin levels were determined via ELISA. Under a long photoperiod, Kiss1 mRNA expression and protein levels exhibited significant diurnal variation, peaking during the light phase ( < 0.05). Pinealectomy disrupted these rhythms, leading to altered testosterone levels. Under a short photoperiod, a marked change in Kiss1 expression and testosterone was also observed, emphasizing photoperiod and melatonin's role in synchronizing reproductive rhythms. These findings highlight the central role of melatonin not just in modulating levels, but also in synchronizing the daily reproductive rhythms necessary for seasonal adaptation. - Source: PubMed

In mammals, FGF21 regulates glucose and lipid metabolism, and energy balance. Fgf21 was recently demonstrated to stimulate feed intake and regulate energy balance in zebrafish. The physiological mechanisms that regulate energy balance are tightly interlinked with reproduction. As a first step to extend our original findings on Fgf21 and energy balance, this research aimed to determine whether Fgf21 regulates the reproductive axis in zebrafish. Using quantitative PCR, we demonstrate the expression of fgf21, its receptor fgfr1c, and co-receptor β-klotho; and FGF21-like immunoreactivity (immunohistochemistry) in the zebrafish ovary and liver cells. These results suggest local production of FGF21 and possible actions within the ovary. Single intraperitoneal (IP) injection of 1, 10, or 100 ng/g bodyweight FGF21 significantly (ANOVA, P < 0.05) upregulated reproductive hormone genes in the brain; gonadotropin-releasing hormone isoforms (sgnrh/cgnrh-III) and kisspeptin (kiss1), and gonads; aromatase (cyp19a1a) and gonadotropin receptors (fshr, lhr) that are critical for reproductive success. FGF21 significantly (ANOVA, P < 0.05) upregulated vitellogenin transcript levels and total vitellogenin concentration (ELISA) in zebrafish liver cells at 1 h and 24 h post-incubation, respectively. Lastly, at 100 ng/mL, FGF21 induced (ANOVA P < 0.05) oocyte maturation in vitro (determined by germinal vesicle breakdown quantification).
