- Background Scaling and root planing (SRP) is the standard nonsurgical treatment for chronic periodontitis; however, complete mechanical debridement may be limited in deep periodontal pockets, prompting interest in adjunctive laser therapy to enhance clinical outcomes. Objective To compare the clinical effectiveness and safety of adjunctive 940-nm diode laser therapy combined with SRP versus SRP alone in patients with chronic periodontitis. Methods This prospective nonrandomized comparative clinical study included 60 patients with Stage II-III, Grade B periodontitis who were allocated to receive either SRP alone (n = 30) or SRP with adjunctive 940-nm diode laser therapy (n = 30) based on participant preference following an informed discussion at a single tertiary care center. Clinical attachment level (CAL) was the primary outcome. Secondary outcomes included probing pocket depth (PPD), plaque index (PI), gingival index (GI), bleeding on probing (BOP), and treatment-related safety. Clinical assessments were performed at baseline and at 4, 12, and 24 weeks by a blinded, calibrated examiner who was not involved in treatment delivery. Within-group comparisons were analyzed using paired t-tests, between-group comparisons using independent t-tests, and longitudinal changes using repeated-measures ANOVA. Multivariable linear regression was used to identify independent predictors of CAL gain. Results Both treatment groups demonstrated significant improvements in all clinical parameters over 24 weeks (p < 0.001). Compared with SRP alone, adjunctive diode laser therapy produced significantly greater PPD reduction (3.58 ± 0.64 vs. 3.22 ± 0.58 mm; p = 0.026) and CAL gain (3.42 ± 0.78 vs. 3.01 ± 0.69 mm; p = 0.034) at 24 weeks. Significant time × treatment interactions were observed for PPD and CAL (p < 0.05). Multivariable regression confirmed adjunctive laser therapy as an independent predictor of CAL gain. No significant between-group differences were observed in PI, GI, BOP, or adverse events. Conclusions Adjunctive 940-nm diode laser therapy was associated with additional clinical benefits over conventional SRP, including enhanced periodontal pocket reduction and attachment gain, without compromising safety. These findings support its use as a complementary nonsurgical treatment modality for appropriately selected patients with chronic periodontitis and provide prospective clinical evidence to guide periodontal practice. - Source: PubMed

The objective of this study is to compare the clinical and radiographic effectiveness of platelet-rich fibrin (PRF) and conventional blood clot scaffolds using standardized cone-beam computed tomography (CBCT)-based assessment and to identify independent predictors of regenerative success. Methods This prospective, non-randomized comparative clinical study was conducted at the Department of Pediatric Dentistry, Pacific Hospital, Udaipur, Rajasthan, India, and included 40 children with non-vital immature permanent maxillary incisors treated with RET using either a blood clot scaffold (n = 20) or a PRF scaffold (n = 20). CBCT-based radiographic analyses were performed at baseline and the 15-month follow-up. The primary outcome was the percentage increase in radiographic root area (RRA), while secondary outcomes included percentage changes in root length, apical diameter, and periapical lesion size. Clinical outcomes were evaluated throughout follow-up. Between-group comparisons were performed using independent t-tests or Mann-Whitney U tests, as appropriate, and multivariable linear regression was used to identify independent predictors of RRA improvement. Results Radiographic analysis included 37 teeth (blood clot, n = 18; PRF, n = 19). The PRF group demonstrated a significantly greater increase in RRA than the blood clot group (19.15 ± 6.22% vs. 9.72 ± 5.18%; mean difference, 9.43%; 95% confidence interval (CI), 5.86-13.00; p < 0.001; Cohen's d = 1.58). PRF also achieved significantly greater reductions in apical diameter (p = 0.028) and periapical lesion size (p < 0.001), whereas the increase in root length did not differ significantly between groups (p = 0.415). Both groups exhibited significant within-group radiographic improvement and high clinical success rates (blood clot: 90%; PRF: 95%). Multivariable regression identified PRF as the strongest independent predictor of RRA improvement (β = 9.43, p < 0.001), while larger baseline periapical lesion size was negatively associated with regenerative outcomes. Conclusions PRF was associated with significantly greater structural root maturation and periapical healing than the conventional blood clot scaffold while maintaining comparable clinical success. Standardized CBCT-based quantitative assessment supports PRF as an effective scaffold for RET and provides robust evidence to inform clinical decision-making. - Source: PubMed

Salivary high-sensitivity C-reactive protein (hs-CRP) has emerged as a promising non-invasive biomarker, but its association with periodontal disease severity remains insufficiently investigated in Indian populations. Objective To evaluate the association between salivary hs-CRP concentrations and the clinical stage of periodontitis in an Indian population and to examine their relationship with established periodontal clinical parameters. Methods This institution-based cross-sectional analytical study included 150 adults equally allocated into periodontally healthy, Stage I-II periodontitis, and Stage III-IV periodontitis groups (n = 50 each). Comprehensive periodontal examinations were performed according to the 2017 World Workshop Classification, and unstimulated whole saliva samples were collected for hs-CRP estimation using a high-sensitivity enzyme-linked immunosorbent assay (ELISA). Group comparisons were performed using one-way analysis of variance(ANOVA) with Bonferroni post hoc analysis. Correlation analyses and multiple linear regression were conducted to evaluate associations between salivary hs-CRP concentrations and clinical periodontal parameters after adjustment for age, sex, and body mass index (BMI). Results Salivary hs-CRP concentrations increased significantly with advancing periodontitis stage (p < 0.001). Significant positive correlations were observed between hs-CRP and periodontal clinical parameters, particularly clinical attachment level and probing pocket depth. Periodontitis stage remained an independent predictor of salivary hs-CRP after multivariable adjustment. Conclusions Salivary hs-CRP concentrations increase progressively with advancing clinical stage of periodontitis and are independently associated with periodontal disease severity. These findings support the use of salivary hs-CRP as a simple, non-invasive adjunctive biomarker for periodontal assessment and provide contemporary evidence using the 2017 World Workshop Classification in an Indian population. Prospective multicenter studies are warranted to validate its clinical utility and integration into multimarker saliva-based diagnostic strategies. - Source: PubMed

TSA treatment has been previously shown to decrease alcohol (ethanol) consumption with dose and sex differences. However, molecular mechanisms underlying TSA's effects on ethanol consumption remain poorly understood. : We collected the nucleus accumbens shell (NAcSh) of HAD1 rats, which has been used to investigate the impact of TSA treatment on ethanol consumption. RNA-seq profiling of NAcSh followed by IPA and GSEA analysis were conducted. : Gene profiling identified differentially expressed genes (DEGs) with sex- and dose-specific effects, with some genes demonstrating high fold change (FC). Males showed significantly increased and decreased expression following 1 mg/kg TSA treatment. In females, expression significantly decreased following 1 mg/kg TSA treatment, whereas expression increased following 2 mg/kg TSA treatment. Pathways centered on and in males, and and estrogen receptor in females were significantly enriched following TSA treatment, with distinct pathways identified at the 1 mg/kg and 2 mg/kg doses. IL1β and β-estradiol are common upstream regulators among all groups. Unexpectedly, some common DEGs between male and female comparisons have opposite responses to the same dose of TSA. GSEA analysis has identified additional gene sets, hallmark genes and microRNAs, and functions including immune response, metabolism, and estrogen response significantly associated with TSA treatment. : This study successfully identified gene expression evidence that TSA treatment is sex- and dose-specific, underscoring the importance of considering both variables in the development of HDAC-targeted therapies for alcohol use disorders. - Source: PubMed

Zou YiHauser Sheketha RRaba Teresa JBell Richard LLai ZhaoLiang Tiebing - Feeding behavior is a key factor influencing growth performance and feed efficiency in poultry, yet its complex phenotypes remain difficult to quantify accurately, limiting the application of large-scale behavioral data in genetic and molecular analyses. In this study, feeding behavior and feed efficiency-related phenotypes were recorded in 829 Tianfu Nonghua ducks using an automated feeding monitoring system integrated with radio frequency identification (RFID) technology. Genetic parameter estimation indicated that the heritability of feeding behavior traits ranged from 0.25 to 0.39. Individuals were classified into high-frequency feeding (HFF) and low-frequency feeding (LFF) groups. Phenotypic analysis showed that ducks in the HFF group had significantly higher residual feed intake (RFI) and feed conversion ratio (FCR) than those in the LFF group. Based on this grouping, transcriptome sequencing was performed on the hypothalamus, pituitary, and liver tissues. A total of 266, 181, and 94 differentially expressed genes (DEGs) were identified in the three tissues, respectively. Enrichment analysis highlighted tissue-specific pathways, including neuroactive ligand-receptor interaction and calcium signaling in the hypothalamus, the Apelin signaling pathway in the pituitary, and steroid hormone biosynthesis in the liver. Appetite-related genes in the hypothalamus (PMCH, HCRT, and CCKAR) and endocrine and metabolic genes in the pituitary and liver (TRH and NEGR1) showed differential expression between feeding strategies. Weighted gene co-expression network analysis further identified hub genes (ITPKB, BAMBI, and BCKDHB) associated with total feeding bouts (TFB). These findings provide new insights into the genetic architecture of feeding behavior traits and the molecular differences associated with divergent feeding patterns in ducks. - Source: PubMed
