- Circular RNAs (circRNAs) have been reported to be closely associated with tumor progression in multiple malignancies. However, the specific mechanism by which circPRKCA influences tumor progression has not been fully elucidated. CircPRKCA is highly expressed in non-small cell lung cancer (NSCLC) tissues and cells. Knockdown of circPRKCA inhibits the malignant behaviors of NSCLC cells. RNA sequencing results revealed that and mRNAs are positively correlated with circPRKCA. Subsequently, we proved that circPRKCA acts as a molecular sponge for miR-200b-3p. Additionally, miR-200b-3p binds to the 3' untranslated regions (3'UTRs) of and mRNAs to promote their degradation. Overexpression of circPRKCA thereby suppresses this degradation process and coun-teracts the tumor-suppressive effects induced by miR-200b-3p. CircPRKCA functions as the sponge of miR-200b-3p, suppressing the / mRNA degradation driven by miR-200b-3p and accelerating NSCLC progression. - Source: PubMed

Zhong HeWang NingZhang HuiChen MinLiao XinHuang Chao - L-theanine (LT), a characteristic amino acid found in tea, possesses diverse biological activities, including antioxidant, growth-promoting, and immune-enhancing effects, and is widely used in supplements. However, the regulatory role of LT in milk synthesis in dairy cows remains unclear. This study aimed to elucidate the effects of LT on milk synthesis and its underlying molecular mechanisms in primary bovine mammary epithelial cells (BMECs). We isolated primary BMECs and investigated the effects of LT treatment (100 µM), utilizing overexpression and knockdown techniques for TAS1R1, FERM domain containing 6 (FRMD6), and mTOR. Results showed that 100 µM LT significantly promoted BMECs proliferation and the synthesis of milk protein and fat. Knockdown of its putative receptor, TAS1R1, reversed these stimulatory effects of LT. Transcriptome sequencing analysis revealed FRMD6 as a key target gene mediating the LT-induced promotion of BMEC proliferation and milk synthesis. Mechanistic studies further indicated that LT enhanced the interaction between FRMD6 and mTOR, promoted mTOR translocation from the cytoplasm to the lysosomal surface, subsequently leading to the activation of the mTORC1 signaling pathway, ultimately promoting milk synthesis. The clinical experiment results on dairy cows show that Rumen-Protected LT can significantly increase the milk production of mid-lactation cows and improve the quality of their milk. In conclusion, our research provides strong preclinical evidence for LT as a potential functional feed additive to promote milk synthesis. - Source: PubMed

Herein, silicon nitride nanoparticles (SiN NPs) were demonstrated to enhance early vascular regeneration and promote new bone formation in vivo. We found that SiN NPs induced autophagy and stimulated angiogenesis after they were uptaken by human umbilical vein endothelial cells (HUVECs). Transcriptome sequencing revealed that the angiogenic effect of SiN NPs was closely associated with modulation of adherens junctions (AJs). Specifically, SiN NPs significantly increased phosphorylation of the core AJs protein VE-cadherin in HUVECs, thereby inducing AJs dissociation-a crucial step in angiogenesis initiation. Furthermore, FRMD6 was identified as the most significantly altered upstream regulator. Knockdown of FRMD6 in endothelial cells restored AJs stability and abolished the SiN NP-mediated pro-angiogenic effects. Local FRMD6 knockdown also impaired the ability of SiN NPs to promote neovascularization and bone regeneration in the rat calvarial defect model. These findings suggest that SiN NPs promote angiogenesis via the FRMD6/VE-cadherin pathway. In the ovary, ECM must dynamically remodel in each cycle under hormonal regulation to control follicle development and produce fertilizable oocytes. Dysregulation of this process may result in aberrant formation of ECM as seen in polycystic ovary syndrome (PCOS) whose pathology includes fibrosis of the ovary and which is a major cause of infertility. PCOS is characterised by hyperandrogenism and, here, we investigate the impact of androgens on fibrosis, cell-ECM interactions and mechanosensing. We report an altered network of gene expression related to the genesis of fibrosis. Preantral follicles from C57BL/6 mice (14-15 days postpartum) were stimulated with dihydrotestosterone (DHT, 10nM) in 24/72 hours culture. Expression of fibrosis-associated genes (Eln; Ctgf; Acta2; Plod2; Hpse) significantly increased with androgen (72 h), as did TGF-β signalling (Tgfb1; Tgfb3). We show a direct connection between androgen and mechanosensing within the ovary, with androgen upregulating the mechanosensitive Hippo pathway (Yap1; Lats1; Lats2; Stk3; Stk4; Frmd6) and downstream targets (Ctgf; Axl; Cyr61). Our results highlight hyperandrogenism as a probable driver of the fibrosis in the polycystic ovary, and emphasise the importance of ECM regulation in follicle development and fertility. - Source: PubMed

Hopkins Thomas I RLerner AvigdorDunlop Iain EFranks StephenHardy Kate - BACKGROUND: Bladder cancer (BC) ranks as the most prevalent malignant neoplasm affecting the urinary system. However, its clinical prevention and treatment continue to face significant challenges. This research aims to investigate the biological roles and regulatory mechanisms of FRMD6-AS2 in BC. METHODS: Tissue samples were collected from 132 BC patients, along with their matched adjacent normal tissues. Quantitative reverse transcription polymerase chain reaction was employed to determine FRMD6-AS2 and miR-1260a expression, and their correlation was assessed using Pearson correlation analysis. The link between FRMD6-AS2 and patient prognosis was analyzed via Kaplan-Meier and Cox regression analyses to identify independent prognostic factors. Cell proliferation, migration, invasion, and apoptosis were determined by employing the cell counting kit-8, Transwell, and Annexin V-fluorescein isothiocyanate/ propidium iodide assays. The direct binding interaction between FRMD6-AS2 and miR-1260a was verified by luciferase reporter and RNA immunoprecipitation assays. RESULTS: FRMD6-AS2 was lowly expressed in BC tissues and cells. Patients with low FRMD6-AS2 expression exhibited poorer overall survival, and it was identified as an independent prognostic factor. Overexpression of FRMD6-AS2 led to the inhibition of BC cell proliferation, migration, and invasion while promoting apoptosis. Mechanistically, FRMD6-AS2 negatively regulated miR-1260a expression. Furthermore, overexpression of miR-1260a partially reversed the suppressive impacts of FRMD6-AS2 on the malignant behaviors of BC cells. CONCLUSION: FRMD6-AS2 exhibits reduced expression in BC and plays tumor-suppressive effects by sponging miR-1260a, suggesting its potential as a prognostic biomarker and therapeutic target for BC. - Source: PubMed
