- The selection of high feed efficiency (FE) animals impacts sustainability and profitability of beef and dairy cattle production systems. An approach to investigate the mechanisms of FE involves analyzing gene expression profile in liver. This study used residual feed intake as a metric of FE to select 10 Gir x Holstein crossbred cows (Girolando F1) divided into high (HE) and low (LE) FE groups. Hepatic biopsies were used for differential gene expression investigation using RNA-seq analyses which revealed 20,787 known genes mapped accordingly to the bovine reference genome. The comparison of HE and LE revealed 149 significantly differentially expressed genes (DEG), 41 up-regulated, and 108 down-regulated in the LE group. Among DEG, some stood out as potential candidate genes, including DLK1, CACNG4, SLC2A12, SLC26A4, DUOX2, and DUOXA2. Functional enrichment analyses showed pathways that potentially influence FE, such as the negative regulation of leukocyte migration, regulation of calcium channel activity, negative regulation of cell migration and adhesion, extracellular matrix (ECM) organization, and thyroid hormone synthesis. ECM composition and immune system roles were also highlighted. Although fetal infection status and disease outcomes vary, the mechanisms are not completely understood. The objective was to assess targeted placental structural and transporter-related gene expression patterns. At day 85 of gestation pregnant pigs were challenged with PRRSV, and at 12 days post maternal infection sows and fetuses were sacrificed, and the placental tissue was collected. Grouping of fetuses was by preservation status and PRRS viral load (VL): control (CTRL, n = 14), viable and low VL fetus (VIA_LVF, n = 15), viable and high VL fetus (VIA_HVF, n = 21), meconium mild and low VL fetus (MECm_LVF, n = 14), meconium mild and high VL fetus (MECm_HVF, n = 14), and meconium severe and high VL fetus (MECs_HVF, n = 13). NanoString was used to evaluate the expression of 86 genes: actin cytoskeleton signaling, arachidonic acid pathway, integrin signaling, intercellular junctions, transporters, and VEGF signaling. Here, we performed a genome-wide association study (GWAS) to identify loci associated with the WML volume. Brain MRI and DNA samples were collected from 9479 participants in the Japan Prospective Studies Collaboration for Aging and Dementia (JPSC-AD). The GWAS confirmed three known WML-associated loci (SH3PXD2A, GFAP, and TRIM47). The lead variant of GFAP was a common missense variant (p.D295N) in East Asians. Meta-GWAS using the publicly available summary statistics of UK Biobank identified one previously unreported locus 6q23.2 (SLC2A12). Integration with expression quantitative trait locus data implied the newly identified locus affects SLC2A12 expression. The effect sizes of 20 lead variants at the WML-associated loci were moderately correlated between JPSC-AD and UK Biobank. These results indicate that the alteration in GFAP protein caused by the common missense variant in East Asians influences the WML volume. - Source: PubMed

Ren Zi-QiWang Ren-DongWang CongRen Xiao-HuiLi Dong-GuoLiu Ya-LingYu Qiu-Hong - Numerous studies over the past few decades have shown that RNAs are multifaceted, multifunctional regulators of most cellular processes, contrary to the initial belief that they only act as mediators for translating DNA into proteins. LncRNAs, which refer to transcripts longer than 200nt and lack the ability to code for proteins, have recently been identified as central regulators of a variety of biochemical and cellular processes, particularly cancer. When they are abnormally expressed, they are closely associated with tumor occurrence, metastasis, and tumor staging. Therefore, through searches on Google Scholar, PubMed, and CNKI, we identified five five recently characterized lncRNAs-Lnc-SLC2A12-10:1, LncRNA BCRT1, lncRNA IGFBP4-1, LncRNA PCNAP1, and LncRNA CDC6-that have been linked to the promotion of cancer cell proliferation, invasion, and metastasis. Consequently, this review encapsulates the existing research and molecular underpinnings of these five newly identified lncRNAs across various types of cancer. It suggests that these novel lncRNAs hold potential as independent biomarkers for clinical diagnosis and prognosis, as well as candidates for therapeutic intervention. In parallel, we discuss the challenges inherent in the research on these five newly discovered lncRNAs and look forward to the avenues for future exploration in this field. - Source: PubMed
