- Global climate change has brought severe challenges to the livestock industry, among which heat stress (HS) acts as a key factor impairing the growth and development of pigs. It has been established that ambient temperature influences body size traits mainly by directly affecting skeletal development, yet the molecular mechanisms by which HS inhibits this process remain largely unclear. The cytoskeleton is critical for mediating cellular morphological adaptation to environmental stimuli, and HS can disturb cellular development by destroying cytoskeletal homeostasis. Previous studies have demonstrated that RFLNA regulates bone development through the cytoskeleton, but whether it alleviates HS-induced chondrocyte damage by modulating the cytoskeleton has not been reported. This study investigated the morphological and genetic basis of body shape variation between Hebao red carp (HB, ) and Yellow River carp (YR, ). Although both subspecies share an identical vertebral count (35), the compressed morphology of HB was attributable to skeletal anomalies, including vertebral shortening and fusion. Genome-wide association and population genetic analyses were performed on F1 and F2 hybrid cohorts to identify loci associated with this phenotype. A total of 231 selective sweep regions were detected across chromosomes A06, A08, A16, B05, and B06, with a prominent locus on chromosome A08 (15.99-16.39 Mb) strongly correlated with body shape traits. Transcriptomic analysis revealed haplotype-dependent expression of within this interval, implicating in axial skeletal patterning. Functional validation using CRISPR/Cas9-mediated knockout of in zebrafish ( ) induced vertebral malformations, including axial shortening, kyphosis, fusion, and a rounded abdominal profile. In this study, we identify WDR60, a component of the dynein-2 complex essential for retrograde intraflagellar transport, as a novel modulator of pancreatic neuroendocrine tumor (Pa-NET) behavior. Transcriptomic analysis of the GEO dataset GSE73338 revealed that WDR60 is significantly upregulated in both primary and metastatic Pa-NETs compared to normal pancreatic islets. Moreover, WDR60 expression is higher in G2 compared to G1 Pa-NETs. Functional analyses in QGP-1 cells following WDR60 silencing demonstrated broad transcriptional reprogramming with enrichment of pathways related to cell adhesion and extracellular matrix (ECM) remodeling. Notably, WDR60 knockdown reduced cell migration and enhanced adhesion without affecting cell viability or proliferation. Among the key upregulated genes were PIK3AP1, RAP1B, and RFLNA, suggesting that WDR60 is involved in regulating PI3K/AKT signaling and cytoskeletal dynamics. To explore potential therapeutic implications, we examined the effects of Ciliobrevin A (HPI-04), an inhibitor of Hedgehog signaling and ciliogenesis. HPI-04 significantly reduced WDR60 expression, impaired cell migration, and increased adhesion. RT-qPCR confirmed overlapping gene expression changes between HPI-04 treatment and WDR60 silencing, although some differences, such as CD164 regulation, suggest WDR60-independent mechanisms. Collectively, these findings identify WDR60 as a critical regulator of cell motility and adhesion in Pa-NETs via ciliary signaling and cytoskeletal remodeling. Despite extensive studies on modern domestic horse origins, the intricate demographic history and genetic signatures underlying pony size remain unexplored. Here, a high-quality genome assembly of the Chinese Debao pony is presented, and 452 qualified individuals from 64 horse breeds worldwide are extensively analyzed. The authors' results reveal the conservation of ancient components in East Asian horses and close relationships between Asian horses and Western pony lineages. Genetic analyses suggest an Asian paternal origin for European pony breeds. These pony-sized horses share close genetic affinities, potentially attributed to their early expansion and adaptation to local environments. In addition, promising cis-regulatory elements influencing horse withers height by regulating genes such as RFLNA and FOXO1 are identified.
