- Pulmonary function (PF) progressively declines with aging. Forced expiratory volume in the first second (FEV1) and forced vital capacity (FVC) are predictors of morbidity of pulmonary and cardiovascular diseases and all-cause mortality. In addition, reduced PF is associated with elevated chronic low-grade systemic inflammation, glucose metabolism, body fatness, and low muscle strength. This study identifies the primary glioblastoma. prognostic genes. Gene expression data in primary glioblastomas with short-term (36 months, N=23) overall survival were downloaded from Gene Expression Omnibus (GSE53733). Limma determined the differentially expressed genes (DEGs) between different groups (|log2 fold change| ≥0.5 and p-value DEG's degree, betweenness, sub-graph and closeness centralities. Long- term/short-term survival-related DEGs were defined as those with increased/decreased expression values and survival time. The following DEGs were identified; 161 between intermediate and short-term glioblastomas, 465 between long-term and short-term and 624 between long-term and intermediate tumors. The common FLRT1 and LINGO1 up-regulated DEGs and common down-regulated C7orf31 were identified in these three DEG sets. PPI networks were established, and VEGFA was the key DEG in each PPI network. The short-term survival-related DEGs were enriched in 3 cancer-related pathways. Moreover, FLRT1 and LINGO1 were long-term survival-related DEGs and C7orf31 and VEGFA were short-term survival DEGs. LINGO1, C7orf31, and VEGFA were confirmed using a further dataset, and we therefore conclude that LINGO1 might be a positive primary glioblastoma prognostic gene and C7orf31 and VEGFA might be negative prognosticators. - Source: PubMed
Liu SXu YZhang S - Obesity is increasing in Africa, but the underlying genetic background largely remains unknown. We assessed existing evidence on genetic determinants of obesity among populations within Africa. MEDLINE and EMBASE were searched and the bibliographies of retrieved articles were examined. Included studies had to report on the association of a genetic marker with obesity indices and the presence/occurrence of obesity/obesity trait. Data were extracted on study design and characteristics, genetic determinants and effect estimates of associations with obesity indices. According to this data, over 300 polymorphisms in 42 genes have been studied in various population groups within Africa mostly through the candidate gene approach. Polymorphisms in genes such as ACE, ADIPOQ, ADRB2, AGRP, AR, CAPN10, CD36, C7orf31, DRD4, FTO, MC3R, MC4R, SGIP1 and LEP were found to be associated with various measures of obesity. Of the 36 polymorphisms previously validated by genome-wide association studies (GWAS) elsewhere, only FTO and MC4R polymorphisms showed significant associations with obesity in black South Africans, Nigerians and Ghanaians. However, these data are insufficient to establish the true nature of genetic susceptibility to obesity in populations within Africa. We hypothesized that centriole proteins have been under-represented in proteomic studies of the centrosome, because of the larger amount of pericentriolar material making up the centrosome. In this study, we have overcome this problem by determining the centriolar proteome of mammalian sperm cells, which have a pair of centrioles but little pericentriolar material. Mass spectrometry of sperm centrioles identifies known components of centrioles and many previously uncharacterized candidate centriole proteins. Assessment of localization of a subset of these candidates in cultured cells identified CCDC113, CCDC96, C4orf47, CCDC38, C7orf31, CCDC146, CCDC81 and CCDC116 as centrosome-associated proteins. We examined the highly conserved protein CCDC113 further and found that it is a component of centriolar satellites, is in a complex with the satellite proteins HAP1 and PCM1, and functions in primary cilium formation. - Source: PubMed
