- Breast cancer is a multifaceted illness impacted by genetic factors as well as environmental influences. While variants in the NUDT17 gene have been associated with cancer biology, their involvement in breast cancer is still inadequately investigated, especially within South Asian populations. This study investigated the association between two NUDT17 polymorphisms, rs9286836 and rs2004659, and breast cancer risk in Bangladeshi women using a case-control design. A total of 240 breast cancer patients and 240 age-, sex-, and BMI-matched healthy controls were enrolled. Genomic DNA was extracted from blood samples, and genotyping was performed using tetra-primer ARMS-PCR for rs9286836 and PCR-RFLP for rs2004659. The statistical methods employed included chi-square tests for genotype distributions and logistic regression to calculate odds ratios (ORs) with 95% confidence intervals (CIs). Genotyping for NUDT17 rs9286836 was successfully completed for all recruited participants. However, for rs2004659, high-quality genotyping data were available for 204 breast cancer cases and 204 controls after quality control procedures. No significant association was observed between rs9286836 and breast cancer risk across the tested genetic models. In contrast, rs2004659 showed a robust protective association with breast cancer, particularly among heterozygous individuals, and this protective effect was consistently observed across the dominant, over-dominant, and allelic models. Haplotype analysis revealed that the AA haplotype was associated with an increased risk of breast cancer, whereas the AG and GA haplotypes were associated with a reduced risk. Expression analysis further demonstrated elevated NUDT17 levels in breast cancer tissues and genotype-dependent expression effects for both variants. These findings suggest a protective role of rs2004659 in breast cancer susceptibility and highlight the potential of NUDT17 polymorphisms as biomarkers in Bangladeshi women. - Source: PubMed

Calmodulin plays a crucial role in plant stress responses as a calcium sensor. However, the functional role of NtCaM13 in drought resistance remains poorly understood. In this study, we found that NtCaM13 overexpression (OE) markedly increased plant resistance to drought, whereas knockout (KO) mutants exhibited heightened susceptibility to water deficit. OE of NtCaM13 reduced drought-induced oxidative damage by increasing antioxidant enzyme activity. Furthermore, transgenic plants overexpressing NtCaM13 exhibited increased antioxidant gene expression, decreased stomatal conductance, and improved water-retention capacity. Nevertheless, the opposite trend was observed in NtCaM13 KO plants. Further RNA-seq analysis revealed that OE of NtCaM13 enhanced the expression of genes involved in stomatal development and closure (ERL1 and SCRM2), cuticle/cell wall reinforcement (LCAO, CYP86A7, PRP1, and XTH9), osmotic regulation (AQP and ProDH2), and antioxidant defense (NUDT17). Conversely, the expression of these genes was diminished in NtCaM13 KO plants. Diphosphoinositol pentakisphosphate kinases (PPIP5Ks) generate the active signaling molecule 1,5-InsP8. PPIP5Ks harbor phosphatase domains that hydrolyze PP-InsPs. Plant and Fungi Atypical Dual Specificity Phosphatases (PFA-DSPs) and NUDIX phosphatases (NUDTs) are also involved in PP-InsP degradation. Here, we analyze the relative contributions of the three different phosphatase families to plant PP-InsP catabolism. We report the biochemical characterization of inositol pyrophosphate phosphatases from Arabidopsis and Marchantia polymorpha. Overexpression of different PFA-DSP and NUDT enzymes affects PP-InsP levels and leads to stunted growth phenotypes in Arabidopsis. nudt17/18/21 knock-out mutants have altered PP-InsP pools and gene expression patterns, but no apparent growth defects. In contrast, Marchantia polymorpha Mppfa-dsp1ge, Mpnudt1ge and Mpvip1ge mutants display severe growth and developmental phenotypes and associated changes in cellular PP-InsP levels. Analysis of Mppfa-dsp1geand Mpvip1ge mutants supports a role for PP-InsPs in Marchantia phosphate signaling, and additional functions in nitrate homeostasis and cell wall biogenesis. Simultaneous elimination of two phosphatase activities enhanced the observed growth phenotypes. Taken together, PPIP5K, PFA-DSP and NUDT inositol pyrophosphate phosphatases regulate growth and development by collectively shaping plant PP-InsP pools. - Source: PubMed
