- This study reports the synthesis and characterization of two target macrocycles (5 and 10) intended to serve as chemosensors for metal cations relevant to biological processes. According to our findings, macrocycle 5 exhibited a marked fluorescence response toward Ag+ and Co2+, exhibiting 75.2% fluorescence quenching with Ag+ and fluorescence enhancement with Co2+ (73.5%). Comprehensive NMR studies provided in-depth insight into the binding mechanisms between the macrocycles and the investigated cations. The analysis revealed that the interaction with Ag+ is strongly influenced by conformational rearrangements of the macrocyclic framework in three-dimensional space, suggesting a dynamic adaptation of the host structure to optimize the coordination. Overall, these results indicate that macrocycle 5 can act as an effective dual chemosensor for metal cations, capable of discriminating between different species through distinct fluorescence responses, and thus represents a promising platform for the development of advanced sensing systems in biologically relevant environments. - Source: PubMed
Larocca MicheleBuonocore MichelaMeo Davide AlfredoGrimaldi ManuelaD'Ursi Anna MariaSoriente Annunziata - In Romania, the HIV epidemic attributed to male-to-male sexual contact is high, due to multiple sociocultural manifestations of stigma toward gay, bisexual, and other men who have sex with men (GBM), which is associated with inadequate HIV prevention. The current analyses describe the results of an intervention aiming to reduce HIV risk and heavy alcohol use, and improve mental health and minority stress-related outcomes, among GBM in Romania. Between 2019 and 2023, 300 Romanian GBM reporting HIV risk and heavy alcohol use were randomized to receive eight 1-h text-based live counseling sessions (the Comunica condition) or self-directed online educational control modules, both informed by the Information-Motivation-Behavior (IMB) model and minority stress theory. Self-reported assessments were conducted at baseline and at 4-, 8-, and 12-month follow-ups; HIV and STI testing was self-administered at baseline and 12-month follow-up. Analyses compared the conditions in terms of their impact on the primary outcome - past-30-day HIV-transmission-risk behavior. Secondary outcomes included mental (e.g., depression symptoms) and behavioral health (e.g., alcohol use) measures. We also examined between-condition change in hypothesized IMB and minority stress mechanisms. The conditions did not significantly differ in any primary or secondary outcomes over time. Both conditions demonstrated decreases in past-30-day HIV-transmission-risk behavior, number of heavy drinking days, depression and anxiety symptoms, and suicidal ideation over the 12-month follow-up period. Education control participants reported significantly greater increases in alcohol- and HIV/AIDS-related knowledge, while Comunica participants reported significantly greater decreases in perceived benefits of condomless sex across the follow-up period. Both conditions showed improvements in self-efficacy for condom use and alcohol use reduction and in all minority stress factors across the follow-up period. As all participants reported similar improvements across outcomes, future research is needed to determine how to best support this stigmatized population's health given the limited population-specific supports. - Source: PubMed

Physicochemical characterization showed improved thermal stability, tunable hydrophilicity, and a porous network suitable for loading and release. In vitro release studies demonstrated sustained cannabinoid delivery over 72 h, following first-order and Fu-Kao kinetic models. The hydrogels exhibited selective antimicrobial activity against , while remaining inactive toward . Cytocompatibility assays with human skin fibroblasts confirmed noncytotoxic behavior over 7 days for all hydrogel formulations tested. Macedonia ChristianCarson Vincent JMorton D HolmesBlakemore Karin JEvans Mark ICrino Peter B - This study evaluated the effects of artichoke bracts silage (ABS) in the finishing diet of beef steers on the microbiological profile, biogenic amines, volatile compounds, and sensory characteristics of dry-aged beef. Forty-eight Holstein × Belgian Blue steers were assigned to three diets: control with wheat straw (C), or diets in which wheat straw was partially replaced with 50 g/kg DM (S1) or 100 g/kg DM (S2) ABS. After 60 days of feeding, bone-in loins were collected and dry-aged for 42 days at 2 °C, 82% relative humidity, and 0.4 m/s air velocity. Longissimus lumborum samples were analyzed at 0, 14, 21, 28, 35, and 42 days. Total mesophilic and psychrotrophic bacterial counts were not affected by diet. Meat from ABS-fed steers showed lower total biogenic amine content from day 28 onward, lower histamine and putrescine concentrations, and delayed cadaverine appearance compared with control meat. Several volatile compounds related to lipid oxidation, amino acid degradation, and dry-aging biochemical development, including aldehydes, ketones, alcohols, and aliphatic hydrocarbons, were higher in control samples than in ABS meat. Dry aging improved tenderness and meaty flavor, whereas control meat developed higher unpleasant odor and taste scores at advanced aging times. Overall sensory appreciation was unaffected by diet. These findings suggest that dietary ABS may modulate biochemical and sensory changes during prolonged dry aging without impairing the overall sensory profile of beef. - Source: PubMed
