- Hyaluronan (HA) is a major glycosaminoglycan of the extracellular matrix that regulates cell migration, signaling, and tissue homeostasis. Its turnover is controlled by coordinated synthesis by HA synthases and degradation by hyaluronidases. Among these hyaluronidases, TMEM2, the only known transmembrane hyaluronidase, plays a unique role in HA degradation at the cell surface; however, the cellular conditions that support its activity remain incompletely understood. To address this, we developed a cell-based HA turnover assay to examine TMEM2-mediated degradation of endogenously synthesized HA, rather than exogenously added, fluorescently labeled HA used in previous studies. Using this system, we show that TMEM2 readily degrades high-molecular weight HA synthesized by co-expressed HAS3. This degradation occurs only when TMEM2 and HAS3 are co-expressed in the same cells (), whereas co-culture of TMEM2-expressing cells with HAS3-expressing cells supports little or no degradation. Interestingly, HA-binding cell surface receptors CD44 and its homolog LYVE-1 promote efficient TMEM2-mediated HA degradation even under conditions, whereas other HA-binding proteins, including TSG-6, layilin, TLR2, RHAMM, and ICAM-1, do not. These findings suggest a spatially regulated mechanism of TMEM2 activity in which capture of HA at the cell surface, mediated by CD44 or LYVE-1, contributes to efficient HA degradation by TMEM2. - Source: PubMed

While mesenchymal stem cells (MSCs) have shown partial benefit in experimental models, direct functional comparison with lineage-committed lymphatic endothelial progenitor cells (LEPCs) remains limited. : Secondary lymphedema was induced in C57BL/6J mice by circumferential tail skin excision with disruption of superficial lymphatics. Animals received intradermal phosphate-buffered saline (PBS), MSCs, or adipose-derived LEPCs on postoperative days 1 and 7. Lymphatic function was longitudinally quantified using IVIS-based near-infrared indocyanine green (ICG) imaging with standardized region-of-interest analysis. Tail diameter was measured serially throughout follow-up as a complementary morphometric parameter. Tissue remodeling was assessed by Picrosirius Red staining and qualitative LYVE-1 immunofluorescence. Statistical analysis incorporated mixed-effects modeling to evaluate treatment group, sex, time, and their interaction terms, with estimation of effect sizes and 95% confidence intervals. : LEPC-treated mice demonstrated higher IVIS signal intensity than MSC- and PBS-treated animals, consistent with improved lymphatic transport at the injury site. Longitudinal tail diameter analysis showed a more favorable temporal profile in the LEPC group relative to controls, with exploratory analysis suggesting potential variations in temporal profiles between sexes that warrant further investigation in larger cohorts. Histologic analysis showed reduced non-tissue/void area and a more favorable qualitative pattern of LYVE-1 staining in LEPC-treated tissue. : LEPC administration enhanced functional and structural recovery in a murine model of secondary lymphedema. Most immunocytochemical studies have been performed with the routinely formalin-fixed and paraffin-embedded tissue. This study aimed to investigate lymphatic and blood vessels immunocytochemically with frozen sections to explore the masked lymphatic and blood vessels in the routinely processed normal tissues. Several normal monkey tissues were frozen in liquid propane, and frozen sections were immunostained for lymphatic and blood vessels using LYVE-1 for lymphatic vessels and von Willebrand factor for blood vessels. In the liver, sinusoids were immunostained panlobularlly for LYVE-1 compared to LYVE-1 immunostaining limited in the pericentral lobe with the routinely processed liver. In the Jejunal villi, lacteals were surrounded by scattered mall venous capillaries. In the large intestines, a few longitudinal lymphatic vessels were detected in lamina propria. Pancreatic islets were surrounded by baskets of rich venous capillaries. Glomerular capillaries were diffusely positive for von Willebrand factor, suggesting leaking capillaries for von Willebrand factor. Prostate had scattered lymphatic and numerous blood vessels in the stroma with nerve bundles in the fibrous capsule. Many lymphatic and blood vessels were immunostained only in frozen sections, which were not immunostained in the routinely processed tissues. Routine processing masks antigen sites of lymphatic and blood vessels. Therefore, immunocytochemical staining with frozen sections is superior to routinely embedded tissues and this immunostaining would further add histopathological information on tumor spread and liver and kidney biopsies. - Source: PubMed
