- TRPA1 channels function as cold sensors and have been implicated in energy metabolism. We investigated the impact of TRPA1 on thermoregulation. mice exhibited body weight loss and increased food intake when transitioning from 30°C to mild cold (14 days at 22°C), indicating increased energy metabolism in response to cold. mice exhibited elevated oxygen consumption, while their core temperature remained unaffected. The BAT proteome in WT mice after 14 days at 22°C showed increased proteins related to glucose utilization and reduced proteins related to lipolysis, whereas mice revealed increased PRDM16 and proteins associated with peroxisomal/mitochondrial β-oxidation. Transcripts of , and were higher in mice at 22°C compared to 30°C, relative to WT mice, with expression uniquely increased in mice. Thermographic images revealed higher interscapular surface temperature in mice exposed to 22°C. Notably, compounds 2 and 3 incorporate an unusual endoperoxide bridge, forging a 7,8-dioxabicyclo[4.2.1]nonane and a 6,7-dioxabicyclo[3.2.2]nonane scaffold, respectively. Their structures were elucidated through comprehensive spectroscopic analyses, single-crystal X-ray diffraction, and quantum chemical calculations. Anti-adipogenic screening revealed that eight members attenuated lipid accumulation in differentiated 3T3-L1 adipocytes, with 2 exhibiting the highest potency. Structure-activity relationships pinpoint an α-substituted acrolein moiety appears to be critical for activity. We examined how S1P-related gene expression in BAT and subcutaneous white adipose tissue (scWAT) varies with acute exercise, chronic exercise, ambient temperature, aging, sex, and UCP1 deficiency. Young and aged wild-type mice, as well as aged UCP1 knockout (UCP1KO) mice, were studied across acute or chronic exercise paradigms and thermal conditions ranging from 4 °C to 30 °C. In BAT, acute exercise was associated with lower , , and expression in young females, whereas aged males showed lower and after exercise. In scWAT, exercise-associated induction of S1P-related transcripts was most evident in aged males. Female BAT also displayed broader temperature- and chronic exercise-associated differences in , , and β-adrenergic receptor transcripts, whereas male BAT showed fewer transcriptional changes. In UCP1KO mice, these exercise-associated patterns were altered or attenuated. Together, these findings identify a context-dependent adipose S1P-related transcriptional program that differs by sex, age, depot, ambient temperature, exercise condition, and UCP1 status. - Source: PubMed

Early detection of the loss of thermogenic adipocytes would allow improved intervention to abrogate childhood obesity, yet reliable markers of human thermogenic adipocytes remain limited. We previously identified expression of (family with sequence similarity 153 member A) in the developing human adipose tissue. Here, we investigated the expression pattern and potential association of with the thermogenic transcriptional program in the adipose tissue of infants, children, and adolescents. We found that adipocyte expression was triggered by beta adrenergic receptor stimulation, a key signal of adipocyte thermogenesis. protein was confined to preadipocyte and adipocyte nuclei, and mRNA expression positively correlated with the mRNA levels of canonical thermogenic adipocyte markers including uncoupling protein 1 (), myogenic differentiation 1 (), type II iodothyronine deiodinase (), transmembrane protein 26 (), Lim Homeobox 8 (), beta adrenergic receptor 2 (), and peroxisome proliferator-activated receptor gamma coactivator 1-alpha (). Importantly, expression was positively associated not only with the expression of individual thermogenic genes but also with their coordinated co-expression.
