- Hepatocellular carcinoma (HCC) is a leading cause of cancer-related mortality, and PD-L1-mediated immune evasion is a major mechanism of tumor immune suppression and an important target of immune checkpoint blockade. Here, we identified RNF166 as an E3 ubiquitin ligase that promotes immune evasion in HCC. RNF166 expression was upregulated in HCC tissues and correlated with PD-L1 levels, whereas RNF166 did not directly affect the proliferation or apoptosis of HCC cells. In immunocompetent mouse models, RNF166 overexpression accelerated tumor growth and suppressed CD8 + T-cell infiltration and cytotoxic function, while these effects were dependent on PD-L1 signaling and were attenuated in immunodeficient settings. Mechanistically, RNF166 physically interacted with c-Jun and promoted K63-linked ubiquitination of c-Jun at K50/K56, which was associated with reduced K48-linked ubiquitination and enhanced c-Jun stability. Stabilized c-Jun bound to the PD-L1 promoter and promoted PD-L1 transcription, thereby linking RNF166-mediated c-Jun stabilization to tumor immune suppression. Upstream, lactate accumulation increased H3K18la enrichment at the RNF166 promoter and contributed to RNF166 transcriptional upregulation. Therapeutically, pharmacological inhibition of c-Jun with T-5224 enhanced the antitumor effect of anti-PD-L1 blockade. Collectively, our findings delineate a lactate-H3K18la-RNF166-c-Jun-PD-L1 axis that promotes immune evasion in HCC and suggest that targeting RNF166 or the RNF166-c-Jun axis may improve PD-1/PD-L1-based immunotherapy. - Source: PubMed

These new data are based on an exceptionally well-preserved specimen from the Campanian of the Dinosaur Park Formation in Alberta, Canada, which provides valuable morphological details previously lacking for this taxon. A detailed morphological analysis was conducted, which revealed a unique combination of plesiomorphic and derived characters. Phylogenetic analyses provide support for the placement of † within the order Rhinopristiformes, a group that includes modern guitarfishes and sawfishes. As the oldest known freshwater-tolerant representative of Rhinopristiformes, † thus offers novel insights into the early ecological diversification of this group. The present study underscores the broader necessity for comprehensive morphological and systematic research on Late Cretaceous batomorphs. Türtscher JuliaJambura Patrick LKriwet Jürgen - The lack of safe, cost-effective, and eco-friendly antioxidant agents to combat oxidative stress-induced chronic diseases remains a significant challenge in biomedical research. Green synthesis offers a sustainable approach for producing bioactive nanoparticles using plant-derived phytochemicals. In this study, calcium oxide nanoparticles (CaO NPs) were synthesized using fruit extract. The formation of nanoparticles was confirmed by UV-Visible spectroscopy, while X-ray diffraction (XRD), transmission electron microscopy (TEM), and Fourier transform infrared spectroscopy (FTIR) revealed crystalline, spherical nanoparticles stabilized by phytochemical capping. The antioxidant activity of the synthesized CaO NPs was evaluated using DPPH, FRAP, ABTS, hydrogen peroxide, and nitric oxide assays at concentrations ranging from 10-50 μg/mL. The nanoparticles exhibited strong, concentration-dependent radical scavenging activity comparable to ascorbic acid. Data shows the potential of S. cumin-mediated CaO NPs as promising candidates for biomedical applications targeting oxidative stress. Thus, we show that -mediated green synthesis can produce structurally stable CaO nanoparticles with significant antioxidant activity, thereby contributing to the development of eco-friendly nanomaterials for oxidative stress-related biomedical applications. - Source: PubMed

Therefore, it is of interest to evaluate the ability of 5% neem, 5% green tea, 5% turmeric, 0.2% chlorhexidine (CHX) and tap water to disinfect Streptococcus mutans-contaminated toothbrushes after 12 hours of soaking. A randomized double-blind crossover study of 40 students (12-15 years) was conducted. All disinfectants significantly reduced bacterial counts compared to baseline, with the greatest reduction observed with 0.2% chlorhexidine, followed by 5% green tea. Thus, data shows that all disinfectants were effective, 0.2% chlorhexidine was the most effective and 5% green tea was a strong herbal alternative. - Source: PubMed
