- Ca-ATPase (CAP) is a key Ca efflux protein in plants. Our previous research suggests that CAPs may play a crucial role in the adaptation of rhododendrons to high calcium environments. However, the evolution, variation, characteristic expression, and subfunctionalization of this gene family in remain unknown. Through the analysis of pan-genomes and pan-transcriptomes, we elucidated the systematic evolution of CAPs in plants, as well as their characteristic expression patterns in . During the evolutionary process from lower to higher plants, CAPs can be divided into six clades and exhibit structural conservation. CAPs have emerged and differentiated in lower plants such as algae, and they have undergone significant amplification in Eudicots plants like rhododendrons. Three species (, , and ) located in the karst province of Guizhou in Southwest China exhibit the highest copies of CAPs, suggesting a strong association between CAP copy number variation and habitat, particularly in high calcium environments. Through multiple transcriptome analyses, we revealed that CAPs are induced under various environmental/developmental conditions (e.g. karst environments, high altitude, early flower development, hormones, etc.). Co-expression network analysis highlighted key members of calcineurin B-like protein (CBL) and CBL-interacting protein kinases (CIPK) that are associated with the high expression of CAPs. Experimental validation demonstrated that CAPb1 and CAPd1 significantly alleviate high calcium stress, and the CAPb1-CIPK1-CBL1 and CAPd1-CIPK2-CBL1 modules can further enhance the alleviation. Here, we implement a multifaceted investigation of the endodermis in lycopsids to shed light on its evolution in roots. We confirmed the presence of an endodermis with Casparian strips (CSs) in two of the three living lineages of lycopsids, while endodermoid layers with polar lignin caps were identified in the Lycopodiaceae. Although lignified tissues, including CSs, can be preserved in fossils, we did not find evidence for either an endodermis or an endodermoid in early fossils spanning the origin of the vascular plant lineage, the divergence of lycophytes and euphyllophytes, or the split of the early lycophytes and lycopsids. Together with evidence from extant lineages, these observations are most consistent with the endodermis having evolved at least twice independently in vascular plants. Expression analysis of homologous genes of the known seed plant regulatory genes SHORT-ROOT (SHR) and SCARECROW (SCR) suggests an ancestral role in defining the boundary between vascular and non-vascular tissues and the independent recruitment in lycopsids and seed plants. - Source: PubMed

Further concerns include survival-time terminology, numerical inconsistencies across tables and figures, competing-risk methods, multiplicity and residual confounding, including sick-quitter bias. The published record requires correction and reconciliation with the underlying data and statistical analyses. Koulaouzidis GeorgeKoulaouzidis Anastasios - Exposure to a traumatic brain injury (TBI) increases risk of developing psychiatric symptoms including depression, anxiety, and post‑traumatic stress disorder (PTSD). However, biological risk and resiliency factors that help explain the heterogeneity in outcomes are limited. Although ~10% of the population was reported to carry natural autoantibodies to the NMDA receptor, it remains unknown whether carrying anti-NMDAR1 autoantibodies modifies risk for psychiatric outcomes after TBI. Plasma anti-NMDAR1 autoantibody levels were quantified in male active-duty service members before a combat deployment. Outcome measures included post-deployment PTSD (Clinician Administered PTSD Scale-IV), depression (Beck Depression Inventory-2) and anxiety symptoms (Beck Anxiety Inventory). Lifetime TBI was associated with increased predicted depression, PTSD and anxiety symptoms after a combat deployment (N = 606 with TBI, N = 419 without TBI). Higher pre-deployment plasma levels of natural anti-NMDAR1 autoantibodies were modestly but significantly associated with lower predicted post-deployment depression (p = 0.0008) and PTSD symptoms (p = 0.0075), but not anxiety, among individuals with TBI. Within the TBI group, high autoantibody group membership (top quartile) lowered predicted post-deployment CAPS-IV and BDI-II scores by 22% ( ~ 4 points) and 25% ( ~ 2 points), respectively and overall prevalence of psychotropic medication use (p = 0.006). High autoantibody group membership predicted lower odds of reporting post‑deployment moderate-severe depression symptoms (BDI-II > 19) (OR = 0.14, 95% CI 0.01-0.69, p = 0.014). Natural anti-NMDAR1 autoantibodies may be a "resiliency" factor for TBI-associated risk for depression and PTSD symptoms, suggesting potential neuroprotective effects. Future studies should determine if anti-NMDAR1 autoantibodies reach the CNS to suppress glutamate excitotoxicity associated with TBI and if these results generalize across different populations. - Source: PubMed
