- The nucleolus is organized around actively transcribed ribosomal RNA genes (rDNA), where high RNA polymerase I (Pol I) activity creates intrinsic susceptibility to replication stress and DNA damage. Here, we identify the DNA translocase RAD54L as a critical regulator of the nucleolar DNA damage response (nDDR) to rDNA double-strand breaks (DSBs) and replication stress. We show that RAD54L localizes to the nucleolus under basal conditions and is recruited to nucleolar caps following CRISPR-Cas9-induced rDNA-DSBs to promote repair. RAD54L loss results in persistent RAD51 foci, increased nucleolar γH2AX, and micronuclei formation, indicating defective resolution of rDNA lesions and genome instability. Under baseline conditions and replication stress induced by the Pol I transcription inhibitor CX-5461, RAD54L limits the accumulation of ssDNA and coordinates nDDR signaling. We further show that rDNA-DSBs induce RNA polymerase II-dependent RNA-DNA hybrids (R-loops) at intergenic rDNA regions, which facilitate nucleolar reorganization and cap formation and repair factor recruitment. Together, these findings establish RAD54L as a key regulator that coordinates replication stress response and rDNA repair, maintaining rDNA stability and genome integrity. - Source: PubMed

Given that most insecticides act via neurotoxic mechanisms, we reasoned that CAP, despite its well-established action on ryanodine receptors (RyRs), may also disrupt nervous system function through additional targets. Insect neurotoxicity is a coordinated process involving neurons and glial cells: voltage-gated sodium (Na) and potassium (K) channels are common insecticide targets, however, their involvement in CAP-induced neurotoxicity has not been established, and glial contributions are poorly understood. We elucidated CAP's multi-target mechanisms by constructing the first single-cell transcriptomic atlas of the H. armigera ventral nerve cord (VNC) using scRNA-seq, validated by whole-cell patch-clamp electrophysiology and RT-qPCR. Results showed that CAP modulated the kinetics of Na channel in a concentration-dependent manner and suppressed K channel currents, confirming both as novel non-canonical targets. scRNA-seq identified VNC cell types, identifying NKCC (perineurial glia-enriched) and UGT39B1 (subperineurial glia-enriched); both were upregulated after 24 h LC CAP exposure. These results indicate that CAP modulates Na and K channels at relatively high concentrations. As potential functional targets, these channels supplement the primary RyR-mediated action of CAP, and imply a partial mechanistic similarity to pyrethroids via regulation of Na channels. Moreover, the VNC atlas facilitated the identification of glial-specific markers (NKCC and UGT39B1), clarifying glial involvement. Collectively, our work advances multi-target understanding of diamide insecticides, provides candidate synergists, and supports "neuron-glial dual-target" strategies for managing H. armigera resistance. - Source: PubMed

Zhang LingnaHou JiayinLi YuxinWei XinghuiLiu JinhongHe Bingjun - We describe the trajectory of a 24-year-old woman with a history of borderline personality disorder (BPD) and complex trauma who presented with significant dissociative episodes. The initial assessment-including a psychiatric interview, the use of standardized measures (PCL-5, CAPS-5), and collateral information gathering-identified complex post-traumatic-stress disorder (PTSD) comorbid with dissociative identity disorder (DIID) and incomplete diagnostic criteria for BPD. Treatment consisted of an integrated psychiatric and psychotherapeutic approach based on the DBT-PTSD model. After just over one year of follow-up and 45 therapy sessions, the patient was in clinical remission from PTSD, while dissociative episodes attributable to DID persisted at a lower intensity. This case illustrates that response to DBT-PTSD may be favorable in patients with comorbid DID. The one-year follow-up highlights the persistence of dissociative episodes alongside sustained clinical remission of PTSD. The patient also shares her perspective on her therapeutic experience. This is, to our knowledge, the first reported case of the results of DBT-PTSD on a person with both complex PTSD and DID, at one year of follow-up after finishing therapy. - Source: PubMed

So far, it heavily depends on geochronological studies of lunar nearside samples but lacks direct sample constraints from the lunar farside. Here, we present 28 single-clast Ar/Ar ages of impact melt rocks returned by the Chang'e-6 mission. They reveal impact events spanning from ∼4.33 to ∼1.13 billion years (Ga). In particular, three melt clasts date to ∼4.16 Ga, consistent with the recent dating of the Apollo basin, and only two ages fall at 4.0 to 3.7 Ga. The presence of older ages (>4.0 Ga) and lack of predominant age around ∼3.9 Ga suggest that the lunar farside bombardment was not dominated by a cataclysmic spike but by a smooth long-term decline punctuated by later impacts. - Source: PubMed

Due to the lack of culturally relevant survivorship care, we aimed to develop a survivorship care toolkit (SCT) that includes a survivorship care roadmap template tailored to Black men to address the unique survivorship care needs of Black prostate cancer survivors (CaPS).
