- Basal cell carcinoma (BCC) rarely metastasizes to other parts of the body. When metastasis occurs, it usually involves regional lymph nodes. BCC lesions located in the head and neck region have been shown to spread to cervical lymph nodes. Here, we present a case of a slow-growing lesion on the scalp that was diagnosed as a case of BCC. Metastasis to the level V cervical lymph nodes was already present at the time of diagnosis. The patient was managed with complete excision of the lesion, along with a neck dissection for level V lymph nodes, and adjuvant chemotherapy with Vismodegib. Molecular pathology results revealed a unique pattern, comprising the fusion of HMGA2-CHMP1A genes and a mutation involving a premature termination codon (PTC) in the CDKN2B gene. The post-operative work-up revealed a hypermetabolic nodule in level IIB lymph which is more likely inflammatory rather than metastatic. However, considering the possibility of metastasis, the patient is being followed up by a multi-disciplinary team (MDT) for evaluation of level IIB cervical lymph nodes. Liang BingyuLi ManyuZhang Lin - Classical swine fever virus (CSFV) infection induces complete mitophagy, which is essential for the clearance of damaged mitochondria. The endosomal sorting complex required for transport (ESCRT) machinery plays a vital role in mediating phagophore closure and autophagosome-lysosome fusion during starvation-induced autophagy. Nevertheless, its involvement in CSFV-induced mitophagy and the underlying mechanisms remain insufficiently understood. Here, we found that the ESCRT-III subunits including CHMP1A, CHMP1B, and CHMP4B, along with the AAA-ATPase VPS4, were actively recruited to autophagosomes during CSFV-induced mitophagy. Consistent with this, depletion of CHMP1A, CHMP1B, CHMP4B or VPS4A disrupted mitophagic flux, impairing both PINK1-PRKN-dependent and -independent pathways. Further investigations revealed that CSFV transiently recruited these subunits to nascent autophagosomes for phagophore sealing during mitophagy. Remarkably, multiple CSFV nonstructural proteins (NSPs) including NS3, NS4B, NS5A and NS5B interacted with these ESCRT key subunits and colocalized on mitophagosomes. In particular, visceral white-nodules disease, caused by , has severely hindered the small yellow croaker () aquaculture industry. However, research on this disease is limited. To address this gap, the present study employed a 100K SNP chip to genotype individuals from an F1 full-sib family, identify single nucleotide polymorphisms (SNPs), and construct a genetic linkage map for this species. A high-density genetic linkage map spanning a total length of 1395.72 cM with an average interval of 0.08 cM distributed across 24 linkage groups was obtained. Employing post-infection survival time as an indicator of disease resistance, 13 disease resistance-related quantitative trait loci (QTLs) were detected, and these regions included 169 genes. Functional enrichment analyses pinpointed 11 candidate disease resistance-related genes. RT-qPCR analysis revealed that the genes of and are significantly differentially expressed in response to infection in spleen and liver tissues, indicating their pivotal functions in disease resistance. In summary, in addition to successfully constructing a high-density genetic linkage map, this study reports the first QTL mapping for visceral white-nodules disease resistance. These results provide insight into the intricate molecular mechanisms underlying disease resistance in the small yellow croaker. - Source: PubMed
