- Canine mammary tumours (CMTs) are the most common malignancies in female dogs and represent a valuable comparative model for human breast cancer. This study aimed to characterise the molecular and functional profiles of three CMT cell lines: CMT-U27, CMT-U131 and CMT-U229, using a zebrafish xenograft model. Gene ex-pression analysis targeted proliferation markers (MKI67, CCNB1, MYBL2, BIRC5, AURKA), receptor signalling (ESR1, PGR, AR, ERBB2, GRB7, EGFR), apoptosis and survival (BCL2, BAG1), and angiogenesis (VEGFA). Zebrafish larvae were xenotransplanted with tumour cells into either the Perivitelline Space or the Duct of Cuvier to assess larval survival and metastatic spread. The CMT-U27 displayed the most aggressive phenotype, with high expression of proliferation-associated genes and the highest metastatic frequency (60%). CMT-U229 exhibited moderate proliferation, accompanied by overexpression of VEGFA. In contrast, CMT-U131 exhibited lower proliferation, correlating with reduced survival in circulation and limited metastasis (<20%). This study provides the first comprehensive molecular characterisation of CMT cell lines in a zebrafish xenograft model. The findings underline the distinct biological profiles of these lines and demonstrate the utility of this platform for testing targeted therapies in veterinary oncology with translational relevance to human breast cancer. - Source: PubMed

Chmielewska-Krzesińska MałgorzataDybalska-Szczepanek Joanna - A pyrogallol-formaldehyde resin (PGR) column was developed for one-step pre-purification of Ge/Ga generator eluates. The resin selectively adsorbs tetravalent cations (Ge, Sn, Ti) with efficiencies of 81.8%, 42.4%, and 29.8% in standards, reducing Sn and Ti to undetectable levels in generator eluates. Formaldehyde leaching was negligible, and cell viability remained 98% ± 1.7%, confirming biocompatibility. Ge breakthrough decreased >20-fold (0.0008 ± 0.0001% to 0.00003 ± 0.00002%). Manual [Ga]Ga-DOTA-TATE labeling yield increased from 58.9 ± 6% to 75.9 ± 1.8% (n = 10). The column is reusable for ≥10 runs. PGR enables high-purity, high-yield Ga radiopharmaceutical synthesis without additional processing time, resolving pH-dependent tetravalent cation interference via colloid mitigation. - Source: PubMed

The ONAWA trial (ClinicalTrials.gov ID: NCT04142892- Registered on January 6, 2021) was a single-arm study evaluating the preoperative effects of onapristone with extended-release formulation (ONA-XR) (50 mg BID for 21 days) in postmenopausal women with Progesterone Receptor-positive (PgR+)/HER2-negative early BC. No complete cell cycle arrest (CCCA) was achieved at day 21, however ONA-XR significantly suppressed tumor proliferation with a Ki-67 geometric mean change of -17.78% (CI95 -39.76% to 12.20%) particularly in tumors with baseline PgR≥90% (-46.21%; CI -63.02% to -21.76%). Immunohistochemistry showed suppression of phospho-Ser294-PgR and p-STAT3, indicating effects on proliferation and endocrine pathways. Treatment also induced a shift to a more endocrine-sensitive phenotype. Of the 10 patients, 60% experienced adverse events, including one grade 3 increase in gamma-glutamyltransferase. In conclusion, ONA-XR showed early tumor proliferation suppression, particularly in high PgR-expressing tumors, supporting its potential for further clinical evaluation. - Source: PubMed
