- Transcriptional regulation of key target genes by BRCA1 is crucial for its diverse functions. Here, we report fatty acid-binding protein 3 (FABP3) as a novel BRCA1-repressible target gene. Functional BRCA1 represses FABP3 promoter activity while mutant BRCA1 breast cancer cell lines show higher endogenous FABP3 expression which was reduced upon introducing wild-type protein. Furthermore, using deletion constructs we identified a critical minimal responsive FABP3 promoter region and confirmed BRCA1 recruitment to this site by ChIP. FABP3 overexpression promoted proliferation, clonogenicity, and migration in breast cancer cells while conferring sensitivity to chemotherapeutic drugs. TCGA analysis correlated high FABP3/BRCA1 expression with reduced overall survival. These findings implicate FABP3 de-repression as a mechanism linking BRCA1 loss to tumor aggressiveness and chemosensitivity. - Source: PubMed

Our objective was to investigate the effects of 2 strategies of nutritional supplementation on physiological and immunological status of early lactating cows. The first strategy consisted of supplementing vitamin E (all-rac-α-tocopheryl acetate) from 3 weeks before calving (3,000 IU/d before and 1,000 IU/d after calving). The other strategy consisted of supplementing plant extracts (Sambucus nigra, Salix alba, Laurus nobilis, Harpagophytum procumbens, Silybum marianum and Arctium lappa, QUALIMILK product from BIODEVAS Laboratoires, QPB plant extracts) from calving (10 g/d). In anticipation of cows being removed due to health issues or changes in calving dates, 51 Holstein dairy cows were assigned to 3 groups of 17 cows with a final target of 15 cows per group. Each group was randomly assigned to one of the 3 compared treatments: control group (final n = 15), vitamin E group (final n = 16), and QPB plant extract group (final n = 14). Their redox and immune status were monitored during the first 12 weeks of lactation, every 4 weeks. In wk 12, malondialdehyde levels in plasma tended to be lower in the vitamin E and QPB plant extract groups than in the control group. The vitamin E and QPB plant extract groups had lower ex vivo plasma CCL4, IL6 and IL8 cytokine levels, lower proportions of blood regulatory neutrophils, and less ROS production by blood neutrophils than the control group. In terms of immune response, the expression of FABP3 in milk mammary epithelial cells was downregulated in multiparous cows supplemented with vitamin E and QPB plant extracts. Therefore, supplementation with either vitamin E before and after calving or QPB plant extracts after calving clearly reduced the ex vivo inflammatory response and enhance a specific ex vivo immune response shown to be involved in the fight against mastitis. - Source: PubMed

Feed intake regulation (REG) is an effective method to modulate energy intake and fat deposition. This study aimed to explore the impact of feed intake and body fat deposition on reproductive performance. A total of 24 Shaziling sows (parity 3-6) were divided into two groups based on their BF on day 30 of gestation, and subsequently randomly assigned to control (CON) and REG groups: (1) BF between 12 and 16 mm (LB-CON, = 6), (2) BF between 12 and 16 mm (LB-IF, = 6), (3) BF between 18 and 22 mm (HB-CON, = 6), and (4) BF between 18 and 22 mm (HB-RF, = 6). Sows in the LB-CON and HB-CON groups were fed a basic diet at a dosage of 1.88 kg/d. The LB-IF and HB-RF groups received REG and were fed a basic diet at dosages of 2.07 kg/d and 1.69 kg/d, respectively. Higher total litter weight and live litter weight were observed in HB-RF sows ( < 0.05). In addition, HB-CON sows displayed higher serum levels of IL-6, TG, and GLU when compared with LB-CON sows ( < 0.05), whereas HB-RF sows exhibited lower levels of these markers than HB-CON sows ( < 0.05). The lipid droplet area fraction was higher in HB-CON sows than in LB-CON sows ( < 0.05), and this fraction was reduced in HB-RF sows compared with HB-CON sows ( < 0.05). The placental level of TC increased in HB-CON sows when compared with LB-CON sows ( < 0.05). The placental levels of T-AOC and GPX increased in HB-RF sows when compared with HB-CON sows ( < 0.05). Moreover, the mRNA expression of FABP3 was higher in HB-CON sows when compared with LB-CON sows ( < 0.05). The mRNA expression of GLUT3 and PlGF increased in HB-RF sows when compared with HB-CON sows ( < 0.05). In addition, HB-RF sows exhibited increased Shannon, Simpson, and ACE indices when compared with HB-CON sows ( < 0.05). The characterization of the fecal microbiota revealed that HB-RF sows displayed a lower ratio of Firmicutes/Bacteroidota and an increased relative abundance of Limosilactobacillus when compared with HB-CON sows ( < 0.05). Collectively, these observations suggested that reduced feed intake in Shaziling sows with high backfat thickness improved reproductive indices, alleviated placental oxidative stress and inflammatory status, elevated placental nutrient transport and angiogenesis-related gene expression, and reshaped fecal microbiota. However, accurately describing how natural product interventions remodel these pathologies across distinct brain regions and cellular microenvironments remains a critical challenge. : We established an integrated multi-omics framework to decode the neuroprotective mechanisms of solanesol (Sol) in an MPTP-induced PD mouse model. We combined single-cell eQTL-based Mendelian randomization (scMR), transcriptomic localization, and virtual knockout analyses to prioritize cell-type-specific regulatory nodes across neuronal, glial, and vascular populations, avoiding the limitations of traditional bulk targeting. In vivo behavioral assays were conducted, alongside orthogonal validation via airflow-assisted desorption electrospray ionization mass spectrometry imaging (AFADESI-MSI) and gene-metabolite co-enrichment analysis, to map regional metabolic networks and structural spatial reprogramming. : Computational prioritization highlighted cell-type-specific regulatory nodes including PRKCB, PRKCE, PDGFRB, and FABP3/5. In vivo, Sol attenuated motor and cognitive deficits and largely restored the highly compartmentalized spatial distributions of striatal dopamine, L-DOPA, and acetylcholine. Crucially, AFADESI-MSI and co-enrichment analysis revealed that Sol specifically reversed MPTP-induced spatial disruptions by rescuing key neuromodulatory metabolites-including cervonoyl ethanolamide, phosphatidylcholine species, taurine, and NADHX-which were tightly coupled to sphingolipid signaling, fatty-acid transport, mitochondrial translation, and cell-adhesion pathways. Conclusions: Sol ameliorates PD-like pathology not through a singular target, but by choreographing a spatially and cellularly compartmentalized restoration of lipid-inflammatory homeostasis. Furthermore, our integrated single-cell and spatial metabolomic blueprint sets a new methodological paradigm for elucidating the precise execution programs of natural neurotherapeutics. - Source: PubMed
