- The engineering of a 3D lymphatic collecting vessel has been of recent interest due to the lack of treatments for lymphatic valve disruptions. While oscillatory shear stress has been shown to regulate lymphatic collecting vessel morphogenesis, its effect in the context of physiologically relevant extracellular matrix remains elusive. In this study, we used a microfluidic device to elucidate the effect of oscillatory shear stress on human lymphatic endothelial cells cultured on dopamine-modified hyaluronic acid or fibronectin. Here, we show that hyaluronic acid can synergistically enhance mechanoresponsive morphological changes, activate cytoskeleton localization, induce nuclear expression of key mechanosensitive markers (PROX1 and FOXC2), amplify mechanotransduction pathways (VE-CAD and mTORC1), and downregulate capillary lymphatic markers (PDPN and LYVE-1). However, molecular determinants governing tumor susceptibility to NK cytotoxicity remain incompletely understood. Here we employ CRISPR activation (CRISPRa) screening to identify cancer cell surface regulators of NK killing. Using a surfaceome-focused library, we screen human and murine cancer cell lines co-cultured with NK cells, identifying known and novel ligands modulating NK cytotoxicity. Screens reveal established factors including CD43 and previously uncharacterized regulators CD44, PDPN, and Siglec-1/CD169. Validation with orthogonal approaches confirm that disruption of these factors alters NK killing susceptibility in vitro and in humanized mouse models. Mechanistically, we find that CD43-mediated NK resistance operates independently of its proposed interaction with Siglec-7, and that targeting CD43 on NK cells or CAR T cells substantially enhances cytotoxic activity against leukemia. These results establish gain-of-function surfaceome screening as a powerful tool for identifying therapeutic targets for NK cell-based immunotherapy. - Source: PubMed

Eerdekens Mariëlle HBoth CharlotteFreitas Rita L - Diabetic peripheral neuropathy (DPN) affects up to 50% of individuals with diabetes mellitus and remains one of the most prevalent and disabling chronic complications of the disease. Approximately one-third develop painful DPN (pDPN), characterized by spontaneous burning pain, electric-shock sensations, allodynia, and hyperalgesia, all of which significantly impair quality of life. Classical mechanisms, including hyperglycemia-driven oxidative stress, mitochondrial dysfunction, microvascular insufficiency, ion-channel dysregulation, and neuroinflammatory activation, explain the structural and functional deterioration of peripheral nerves but do not fully account for the emergence of painful phenotypes. In this narrative review, we aim to provide an overview of the current understanding of pDPN, focusing on how peripheral metabolic and neuroimmune disturbances interact with central nervous system plasticity to produce chronic pain, and highlight emerging evidence that mechanistic endotypes may guide more precise, individualized therapies. - Source: PubMed
