The objective of this study was to use homozygosity (ROH) and supplementary methods (F, iHS, xp-EHH, Rsb) to identify selection signatures and inbreeding pattern of the indigenous Tswana goat. A total of 216 goats were used, that is, Tswana (n = 114) from three agroecological regions in Botswana and Boer (n = 102), a reference population from South Africa. After quality control, 216 animals and 49 732 single-nucleotide polymorphisms were available for analysis. Tswana goats exhibited an average ROH length of 2.20 Mb and 85.71 ROH per goat, while Boer goats had longer, fewer ROH, averaging 155.96 Mb and 3.14 ROH per goat. Tswana goats had a lower inbreeding coefficient (F = 0.08) compared to Boer goats (F = 0.13). Significant ROH hotspots were found on chromosomes 12, 6, and 5 in Tswana goats and on chromosomes 24, 14, 9, 8, and 6 in Boer goats, with 27 annotated genes identified. Multiple selection signature detection methods detected genes such as PUS7L, ADAMTS20, TWF1, PRICKLE1 YAF2 and GXYLTI. Key genes associated with reproductive fitness (ATP12A, RNF17), immune response (IL17D, PARP4), coat colour variation (ADAMTS20), and milk synthesis (TWF1) were highlighted in Tswana goats, reflecting adaptive responses to environmental pressures. The study provides insights into the genetic adaptations and historical breeding of Tswana goat. This knowledge is crucial for implementing effective conservation strategies and enhancing the resilience of indigenous goat populations. By prioritising the genetic study of these goats, Botswana can ensure the sustainability of its unique livestock resources, promoting both food security and rural development in the region. - Source: PubMed

We leveraged logistic regression models to develop and validate polygenic prediction models to estimate the risk of persistent PN symptoms in a training cohort and validation cohort taking clinical risk factors into account. Based on 337 whole-exome sequenced ESBCS two of five prediction models for individual PN symptoms obtained AUC results above 60% when validated. Using the model for numbness in feet (35 SNVs) in the test cohort, 73% survivors were correctly predicted. For tingling in feet (55 SNVs) 70% were correctly predicted. Both models included SNVs from the ADAMTS20, APT6V0A2, CCDC88C, CYP2C8, EPHA5, NR1H3, PSKH2/APTV0D2, and SCN10A genes. For cramps in feet, difficulty climbing stairs and difficulty opening a jar the validation was unsuccessful. Polygenic prediction models including clinical risk factors can estimate the risk of persistent taxane-induced numbness in feet and tingling in feet in ESBCS. - Source: PubMed

Engvall KristinaUvdal HannaBjörn NiclasÅvall-Lundqvist ElisabethGréen Henrik - The objective of this study was to elaborate Doppler ultrasonographic scan, genetic resistance and serum profile of markers associated with endometritis susceptibility in Egyptian buffalo-cows. The enrolled animals were designed as; twenty five apparently healthy buffalo-cows considered as a control group and twenty five infected buffalo with endometritis. There were significant (p < 0.05) increased of cervical diameter, endometrium thickness, uterine horn diameter, TAMEAN, TAMAX and blood flow through middle uterine artery with significant decrease of PI and RI values in endometritis buffalo-cows. Gene expression levels were considerably higher in endometritis-affected buffaloes than in resistant ones for the genes A2M, ADAMTS20, KCNT2, MAP3K4, MAPK14, FKBP5, FCAMR, TLR2, IRAK3, CCl2, EPHA4, and iNOS. The RXFP1, NDUFS5, TGF-β, SOD3, CAT, and GPX genes were expressed at substantially lower levels in endometritis-affected buffaloes. The PCR-DNA sequence verdicts of healthy and affected buffaloes revealed differences in the SNPs in the amplified DNA bases related to endometritis for the investigated genes. However, MAP3K4 elicited a monomorphic pattern. There was a significant decrease of red blood cells (RBCs) count, Hb and packed cell volume (PCV) with neutrophilia, lymphocytosis and monocytosis in endometritis group compared with healthy ones. The serum levels of Hp, SAA, Cp, IL-6, IL-10, TNF-α, NO and MDA were significantly (P˂0.05) increased, along with reduction of CAT, GPx, SOD and TAC in buffalo-cows with endometritis compared to healthy ones. The variability of Doppler ultrasonographic scan and studied genes alongside alterations in the serum profile of investigated markers could be a reference guide for limiting buffalo endometritis through selective breeding of natural resistant animals. - Source: PubMed
