- Herein we describe unusual or rare morphologic features in two high-grade uterine sarcomas with novel or uncommon genetic alterations, to bring attention to their morphologic diversity, diagnostic challenges, and potential therapeutic implications. We report an uterine sarcoma with a FGFR1::TACC1 fusion, the third reported case in the literature, exhibiting epithelioid morphology not previously documented and negativity for S100 and CD34 immunohistochemistry (IHC). We also report an uterine sarcoma with a novel ACOT7::ERBB4 fusion, which displayed spindle cell morphology and osteosarcomatous differentiation. S100 and SOX10 IHC were both negative. Molecular testing was highly informative in reaching a diagnosis in both cases, due to the unusual morphology and limited immunohistochemical support. Clinical follow-up revealed aggressive behavior in both cases, demonstrated by rapid disease progression and metastatic spread. These two cases expand on the known histologic and molecular spectrum of uterine mesenchymal neoplasms with tyrosine kinase receptor gene fusions. Recognition of these entities is important for appropriate classification, prognostication, and identification of patients eligible for targeted therapies such as TRK or FGFR inhibitors. - Source: PubMed

We investigated the protein networks of calmodulin (CaM), a striatin (STRN)-binding protein implicated in DbCM, and compared them with the STRN network from the same tissues. Using CaM as bait, we precipitated protein clusters from left ventricles (LVs) of diabetic rat hearts at 8 and 24 weeks post-streptozotocin (post-STZ). Diabetic rats exhibited pathological remodeling, evidenced by increased heart-to-body weight ratio, β-MHC protein, and ANF mRNA expression. Western blotting showed elevated STRN, but not PP2A-A or -C subunits, in chronic stages. Proteomic analysis at 24 weeks revealed 49 differentially interacting proteins (DIPs) with CaM: 18 enhanced and 31 diminished in diabetic LVs. Functional annotation highlighted the recruitment of proteins in metabolic pathways (fatty acid elongation and lipid metabolism) and PPAR signaling, alongside reduced interactions in lipid response, glucocorticoid response, calcium signaling, and amino acid biosynthesis. Comparative analysis of CaM and STRN interactomes converged to 231 proteins implicated mainly in metabolic processes. Out of these, three proteins (Hnrnpm, Acot7, and Gsta3) surfaced as regulators of metabolic, oxidative, and post-transcriptional remodeling. These findings reveal novel functional roles for the CaM/STRN complex in DbCM and identify a shared signaling hub that may guide therapeutic strategies for diabetes-associated cardiac deterioration. - Source: PubMed

Gaining insight into these mechanisms is vital for improving production efficiency and guiding molecular breeding in this economically important breed. Although previous studies have described the phenotypic characteristics of Huanghuai sheep, the genetic regulatory networks controlling muscle growth and meat quality at different developmental stages remain unclear. No thorough analysis of growth traits and transcriptomic variations across key age points has been conducted. Therefore, in this study, we aimed to evaluate how growth stage influences muscle development, carcass characteristics, and meat quality in Huanghuai sheep by integrating phenotypic characterization with transcriptomic profiling to identify key genes and molecular pathways underlying these economically important traits throughout development. Sixty Huanghuai sheep were assigned to three groups (twenty per group) representing key developmental stages (3, 9, and 18 months of age). Carcass traits and meat quality were evaluated. RNA sequencing of the muscle was performed to identify differentially expressed genes (DEGs), followed by bioinformatics analysis and experimental validation. The results indicated that the 9-month-old sheep presented a favorable balance of dressing percentage and intramuscular unsaturated fatty acid content, while those aged 18 months old exhibited the highest dressing percentage (61.23%). Transcriptome analysis identified 1395 DEGs ( < 0.05 and |log2FC| > 1) and enrichment analysis revealed key pathways involved in thyroid hormone synthesis, skeletal muscle satellite cell proliferation, and skeletal muscle tissue growth. Several candidate genes for muscle development (e.g., , , ) and meat quality (e.g., , , ) were identified and validated. Their expression patterns showed significant correlations between critical growth performance and fatty acid composition metrics. These findings provide novel insights into the molecular networks regulating economically important traits in Huanghuai sheep, offering valuable targets for future molecular breeding programs aimed at enhancing productivity and meat quality. - Source: PubMed
