- It was long believed that the eukaryotic RNA was primarily capped with 7-methylguanosine (mG). However, the expanding field of noncanonical RNA caps recently revealed the presence of other structures (nicotinamide adenine dinucleotide, flavin adenine dinucleotide, dinucleoside polyphosphates) at the 5' end of various mRNAs and noncoding RNAs. We report here on the detection and quantification of a new type of RNA cap-diadenosine diphosphate (ApA) and adenosine guanosine diphosphate (ApG)-on mammalian RNA. We have established a liquid chromatography-mass spectrometry (LC-MS) method to measure ApN-RNA levels and observed its increase under oxidative stress. To identify ApN-capped RNAs, we have developed a sequencing method (ApN-RNA Seq) by leveraging the ability of truncated T4 RNA Ligase 2 to selectively ligate ApN-capped RNAs to an RNA adaptor. We applied this method to human short RNA and identified a subset of tRNA-derived species as candidate ApN-capped RNAs. Among these, ApN-RNA Seq revealed previously uncharacterized 3' tRNA fragments consistent with ApA capping at their 5' ends. The results revealed weak chloroplast differentiation among , and var. However, chloroplast variation exhibited a strong geographic signal across the studied populations. The observed chloroplast variation primarily reflected geographic structuring rather than clear differentiation among these closely related taxa. In contrast, showed distinct chloroplast haplotypes not shared with the other taxa. These findings demonstrate that the developed chloroplast CAPS marker system is effective for detecting chloroplast haplotype variation but has limited discriminatory power among closely related wild taxa. While pharmacological and psychotherapeutic interventions remain central to treatment, peer support has emerged as a promising complementary approach that fosters agency, psychosocial recovery, and affective connection. This study explored the perceived effectiveness of Peer Support Groups (also known as Peer Support Circles or CAPs in Spanish)-a pioneering, community-based intervention in Chile for individuals diagnosed with bipolar disorder (BD). The research examined participants' experiences, focusing on relational benefits, challenges, and group dynamics. A qualitative design was used. In-depth semi-structured interviews were conducted with 17 participants (7 peer supporters and 10 peer participants). Data were analyzed using Constructivist Grounded Theory (CGT) and intersubjective validation was performed within the research team. Three main analytical categories were identified: (1) Benefits of participating in the CAPs, (2) Conditions that enable CAPs' effective functioning, and (3) Challenges and limitations CAPs face. The central phenomenon that emerged was the CAPs' capacity to lower the barrier of isolation commonly experienced by people with BD through relational safety and peer-driven meaning-making. CAPs operate not only as spaces for support and psychoeducation but also as platforms for identity reconstruction and community reintegration. Findings highlight the value of peer-led models for advancing recovery-oriented care and the importance of institutional support to sustain them and maximize their impact. - Source: PubMed

The tool estimates the effects of local actions tackling greenhouse gas (GHG) emissions associated with two sectors, namely residential buildings and transport, on air quality. It estimates emission changes associated with GHG mitigation actions described in city climate action plans (CAPs) for five key air pollutants, namely fine particulate matter (PM ), nitrogen oxides (NOx), sulphur dioxide (SO ), ammonia (NH ) and volatile organic compounds (VOC). The analysis applies the tool to evaluate potential impacts of local climate action for 494 cities on ambient air pollutant emissions, based on 531 CAPs submitted to the Covenant of Mayors for Climate and Energy (CoM) initiative. The results demonstrate that GHG mitigation actions are often synergic with air pollution control strategies, offering significant air quality co-benefits. The magnitude and type of benefits depend on the sector and pollutant considered. Nevertheless, some actions may pose risks for air quality. There are risks of increasing, for example, ammonia emissions through a transport fuel switch and, more significantly, fine particulate matter emissions if solid biomass for heating in the residential sector is promoted. Therefore, context-specific data and case-by-case tailored analyses are needed to support the adequate integration of GHG mitigation and air quality policies at city level, avoiding potential trade-offs and maximizing synergies. - Source: PubMed
