Abdul Ammer Hasan ZainabHamad Hanaa Kamil - Preeclampsia is a hypertensive and multisystemic disorder characterized by mild to severe hypertension during the second half of pregnancy. Circulating microribonucleic acids (miRNAs) have been identified as potential biomarkers for its diagnosis and prognosis. In this study, we compared global miRNA expression profiling in serum samples obtained from pregnant women belonging to three groups: women with severe preeclampsia pregnancies, mild preeclampsia pregnancies, and healthy pregnancies using the NanoString nCounter platform. We found that 26 miRNAs exhibited significant differential expressions between the three groups. Based on predefined selection criteria, miR-604 and miR-423-5p were initially identified as up-regulated, whereas miR-3615, miR-99b-5p, and miR-4443 were down-regulated by NanoString profiling. These candidate miRNAs were subsequently selected for qRT-PCR validation. qRT-PCR analysis confirmed that miR-99b-5p, miR-4443, and miR-604 were consistently down-regulated in both mild and severe preeclampsia compared with healthy pregnancies. miR-3615 demonstrated stage-dependent variation, with significant downregulation in mild preeclampsia and a relative increase in severe cases. Notably, only miR-423-5p exhibited the most pronounced dysregulation, showing slight reduction in mild preeclampsia but a marked up-regulation in severe preeclampsia, consistent with the NanoString results. These findings suggest that miR-423-5p may serve as a potential circulating biomarker associated with disease severity in preeclampsia. - Source: PubMed

Phupong VorapongChuaypen NatthayaMingmongkol SupawadeeTantbirojn PatouLertkhachonsuk Ruangsak - In 2017 and 2018, studies were conducted in Iowa, US across three environments with a history of greater than expected corn rootworm injury i.e., greater than one node of injury to (US EPA 2009) and one environment without rootworm injury, and at different growth stages, to determine the expression levels of mCry3A and eCry3.1Ab transgenes in the roots of different Bt corn hybrids namely the molecular stack (MZIR098), breeding stack (MIR604 × 5307), 5307 and MIR604. ELISA results showed that expressions of both mCry3A and eCry3.1Ab transgenes, were higher in the MZIR098 molecular stack and breeding stack (MIR604 × 5307), than the MIR604 and 5307 at V3 and VT growth stages over both years. To protect the roots from feeding damage by the corn rootworm larvae, expression of the transgenes must be high at the V3 growth stage. The expression of the transgene was significantly impacted by the stage of plant growth while the environments with greater than expected corn rootworm injury did not impact expression of the transgenes. It was found that the expression of mCry3A and eCry3.1Ab transgenes were high at the V3 plant growth stage compared to the VT growth stage. Stacking two or more genes together in the same plant such as the molecular and breeding stacks have the potential to protect roots in environments with higher-than-expected damage and slow down the evolution of resistance in field populations of rootworms. - Source: PubMed

Increasing evidence indicates that circular RNAs (circRNAs) play multifaceted roles in cellular processes in human cancers. Here, we demonstrated that circFNTA high expression increases the proliferation, metastasis, and epithelial-mesenchymal transition process and tumorigenicity of GC cells. First, we found that circFNTA was upregulated in GC cells and tissues, and the high circFNTA levels were positively associated with the poor prognosis in GC patients. Using luciferase reporter and RNA-pull down assays, we elucidated that circFNTA sponged two microRNAs, miR-604 and miR-647. In addition, the proliferation and metastatic ability of GC cell reduction caused by silencing circFNTA was hindered by inhibitors of miR-604 and miR-647. Moreover, SCN8A was predicted by miRDB as a common target gene of miR-604 and miR-647, which was then verified by the luciferase reporter assay. Knockdown of circFNTA causes messenger RNA and protein levels in SCN8A to be downregulated in GC cells. However, this effect was overturned by cotransfection miR-604 and miR-647. Also, we identified that SCN8A was downregulated in GC tissues, which was positively correlated with circFNTA expression. In rescue experiments, the attenuated cell proliferation and metastatic ability caused by circFNTA knockdown was reversed by miR-604 and miR-647 inhibitors and SCN8A overexpression. Collectively, our findings suggest an oncogenic role of circFNTA in GC progression and elucidate that circFNTA exerts its function by modulating the miR-604/miR-647/SCN8A axis. - Source: PubMed
