Herein, we analyzed the binding efficiency and binding dynamics of SARS-CoV-2 virus-like particles (VLPs), mediated by BSG and DPP4 at the single-particle level, by combining super-resolution direct stochastic optical reconstruction microscopy (dSTORM) and single-molecule force spectroscopy (SMFS). dSTORM imaging demonstrated that DPP4 exhibits higher surface expression levels than BSG on HepG2 cells. However, SARS-CoV-2 VLPs display a distinct preference for BSG-dependent cellular entry. Using SMFS, we further probed the binding dynamics between the SARS-CoV-2 spike (S) protein receptor-binding domain (RBD) and these two receptors. The results demonstrated that RBD-BSG exhibits greater stability and a more rapid formation rate compared to RBD-DPP4. Subsequently, a real-time force tracing technique was used to show that BSG-mediated VLP entry requires lower force and faster speed compared with DPP4-mediated entry. In particular, we confirmed that the structural integrity of intramolecular disulfide bonds within the RBD is indispensable for mediating high-affinity receptor binding. These findings elucidate the distinct receptor-mediated endocytic dynamic mechanisms underlying SARS-CoV-2 entry mediated by BSG and DPP4, expanding our understanding of viral invasion beyond the ACE2 pathway and providing insights into potential intervention strategies targeting viral infection. - Source: PubMed

Degli Esposti MichelleSteele BridgetBach-Mortensen Anders MaltheGoodair Benjamin - Extracts from untreated (fresh) and treated (oven-dried and ground) BSG, the most abundant residue from the brewing industry, were investigated through UHPLC-MS/MS metabolomic analysis to evaluate the effect of biomass pretreatment and extraction conditions on their chemical composition. Three extraction conditions were applied to each starting material, and the identified compounds were grouped into eight classes based on their chemical structure. The accumulated MS signal intensity of each class of compounds was then compared across the starting materials and the extraction conditions. The results showed that for some compound groups (e.g., sugars, amino acids, and phenolic compounds) drying was the main discriminating factor, while for others (lipids and hordatines) the determining factor was the extraction solvent. In addition, antioxidant capacity assays revealed that extracts obtained from untreated BSG with 40% hydroethanolic solvent displayed the highest activity. Overall, our data indicate that the energy- and time-consuming drying/stabilization pre-treatment step may be avoided. Given the predictable availability of BSG in both quantity and timing this is possible, although confirmation studies at pilot and industrial scale are needed. - Source: PubMed

This is partly due to lack of a comprehensive measurement-instrument to assess belief in stages. Thus, the current study sought to develop such an instrument. Sixty-four items were generated by experts and subjected to exploratory and confirmatory factor analysis. This culminated in the 19-item Belief in Stages of Grief Scale (BSG), consisting of five factors (Positive, Anxious, Rigid, Passive, and Prescriptive Endorsement). Convergent validity analyses indicated that belief in stages may not be associated with adverse outcomes. However, limitations of this research, including the use of a predominantly female and well-educated convenience sample, highlight the necessity of further investigation. The current research paves the way for future research into the impact of stage-thinking through the development of the BSG and examination of the relationship between belief in stages and health outcomes. - Source: PubMed
